Cloning, Expression and Bioinformatical Analysis of OsNRAMP7 Gene in Rice

摘要 自然抗性相关巨噬细胞蛋白(natural resistance-associated macrophage proteins,NRAMPs)是一类在结构和功能上保守的大家族,目前已在水稻(Oryza sativa)中鉴定出7个基因(OsNRAMP1~OsNRAMP7),它们在金属离子的吸收、转运和分配等方面发挥着重要作用,但迄今为止,OsNRAMP7功能尚不清楚。本研究克隆了OsNRAMP7基因的全长CDS序列,长度为1626 bp,编码541个氨基酸,位于水稻第12号染色体,由4个外显子和3个内含子组成。生物信息学分析表明Os NRAMP7为稳定疏水性蛋白,存在11个跨膜结构域和多个磷酸化位点,预测定位于细胞质膜。表达模式分析表明OsNRAMP7主要在根、茎和幼穗中表达,响应多种植物激素,受到干旱和NaCl胁迫诱导。本研究结果为进一步研究OsNRAMP7的生物学功能提供了基础资料。 Natural resistance-associated macrophage proteins(NRAMPs) constitute a large family with conserved structure and function. Seven genes(OsNRAMP1~OsNRAMP7) have been identified in rice, which play an important role in the uptake, transport, and distribution of metal ions. However, until now, the function of OsNRAMP7 is unclear. In order to understand the biological function of OsNRAMP7, this study cloned the full length CDS sequence of the OsNRAMP7 gene, which is 1 626 bp in length and encodes 541 amino acids. It is located on rice chromosome 12 and consists of 4 exons and 3 introns. Bioinformatics analysis shows that OsNRAMP7 remain a stable hydrophobic protein with 11 transmembrane domains and multiple phosphorylation sites. It is predicted to localize to the plasma membrane. Analysis of expression patterns showed that OsNRAMP7 is mainly expressed in roots, stems and young spikes, responds to a variety of phytohormones and is induced by drought and NaCl stress. This study provides basic data for further studying the biological function of OsNRAMP7.
