摘要
为了探索木本花卉圆锥绣球的组培快繁体系的建立,本研究以‘北极熊’品种的幼嫩的茎尖和带腋芽的茎段为外植体,探索了适宜的外植体的采集时间、预处理及消毒方法、适宜的启动培养、增殖培养、生根培养基及培养方法。结果表明:适宜的茎段外植体采集时间为4月份;预处理及消毒方法为:将枝条先用75%百菌清1000倍液浸泡约10 min后冲洗,再用洗衣粉溶液浸泡约5 min,然后流水冲洗1~1.5 h,然后在超净工作台内,用75%酒精浸泡消毒60 s,无菌水冲洗3~5遍后,再用0.1%~0.2%HgCl2消毒(4+4)min,然后用无菌水反复冲洗3~5次,其成活率达76.7%。启动培养的最佳配方为MS+6-BA0.5 mg/L+NAA 0.06 mg/L,诱导率可达84.2%。较适宜的增殖配方为:MS+6-BA 2.0 mg/L+NAA 0.5 mg/L,附加蔗糖30 g/L,增殖系数达6.0。适宜的生根培养基为:1/2MS+IBA 1.0 mg/L+NAA 0.2 mg/L,附加蔗糖20 g/L,生根率达83.3%,平均根长2.7 cm。本研究结果对圆锥绣球‘北极熊’的组培快繁提供了技术支持。
In order to explore the establishment of tissue culture and rapid propagation system of woody flower of Hydrangea paniculate, the young stem tip and the stem segment with axillary buds of ’polar bear’ were used as explants in this study. And the appropriate collection time, pretreatment and disinfection methods, suitable start-up culture, proliferation culture, rooting culture mediums and methods of explants were explored. The results showed that the appropriate time for stem explant collection was April, and the pretreatment and disinfection methods were as follows: soaked the branches in 1 000 fold 75% chlorothalonil solution for about 10 min, then washed them in washing powder solution for about 5 minutes, then washed them in running water for 1~1.5 h, then soaked them in 75% alcohol for 60 s, washed them in sterile water for 3~5 times, and then disinfected them in 0.1%~0.2%HgCl2(4+4) min, the survival rate was 76.7%. MS+6-BA 0.5 mg/L +NAA 0.06 mg/L was the best formulation for start-up culture, and the induction rate was 84.2%. The suitable formula of proliferation culture was MS +6-BA 2.0 mg/L+NAA 0.5 mg/L, additional sucrose 30 g/L, and the proliferation coefficient was 6.0. The suitable rooting medium was 1/2 MS+IBA 1.0 mg/L+NAA 0.2 mg/L, additional sucrose 20 g/L, the rooting rate was 83.3%,the average root length was 2.7 cm. This study provided technical support for tissue culture and rapid propagation of Hydrngea paniculate ’polar bear’.
