潜阳育阴颗粒对自发性高血压大鼠肾纤维化的干预作用 被引量:3

Intervention of Qian Yang Yu Yin Granule on Renal Fibrosis in Spontaneously Hypertensive Rats

摘要 目的从肾脏细胞外基质成分的积聚和降解失衡的角度,探讨潜阳育阴颗粒对自发性高血压大鼠肾纤维化的干预作用。方法 WKY大鼠为正常对照组,SHR大鼠随机分为3组,模型组(SHR)、缬沙坦组(SHR+Valsartan)和潜阳育阴颗粒组(SHR+QYYY),以上每组大鼠8只,灌胃给药,每日1次,连续8周。取大鼠血液、肾脏组织做以下检测:①大鼠血压检测,大鼠肾脏组织形态学HE染色,纤维化程度Masson染色;②大鼠肾纤维化指标血清层黏蛋白(Laminnin)、透明质酸酶(HAase)、Ⅲ型前胶原(Precollagen Ⅲ)和Ⅳ型胶原(Collagen Ⅳ)含量测定。结果 ①潜阳育阴颗粒可降低SHR大鼠收缩压,舒张压,平均动脉压水平;②潜阳育阴颗粒通过抑制炎症细胞在肾间质的积聚,改善肾脏的病理情况,减少胶原纤维沉积,减轻肾纤维化程度;③潜阳育阴颗粒可降低SHR大鼠血清纤维化指标的含量。结论潜阳育阴颗粒可以有效降低血压,降低血清肾纤维化指标的含量,减轻肾纤维化程度。 Objective From the perspective of the accumulation and degradation of extracellular matrix components in the kidney, to explore the intervention effect of Qian Yang Yu Yin Granules(QYYYG) on renal fibrosis in spontaneously hypertensive rats.Methods WKY rats were the normal control group. SHR rats are randomly divided into 3 groups, model group(SHR), valsartan group(SHR+ Valsartan), and QYYY group(SHR+QYYY), each of the above groups has 8 rats intragastric administration once a day for 8 weeks. Take rat blood and kidney tissue for the following tests, 1 Determination of rat blood pressure, rat kidney HE staining, Masson staining;2 Determination of serum laminin(Laminin), hyaluronidase(HAase), type Ⅲ procollagen(Procollagen Ⅲ), and type Ⅳ collagen(Collagen Ⅳ) renal fibrosis markers in rats.Results 1 QYYYG can reduce the levels of SBP, DBP and MAP in SHR;2 QYYYG improves the pathological condition of the kidney by reducing the accumulation of inflammatory cells in the renal interstitium, reduces the deposition of collagen fibers, and reduces the degree of renal fibrosis;3 QYYYG can reduce the serum Laminnin, HAase, Procollagen Ⅲ and Collagen Ⅳ levels in SHR.Conclusion QYYYG can effectively reduce blood pressure, reduce the content of serum fibrosis markers and reduce the degree of renal fibrosis.
