天冬多糖协同HIF1αRNAi在缺氧环境下抑制肝癌血管生成的研究 被引量:3

摘要 目的研究天冬多糖联合HIF1αRNAi在缺氧环境下对肝癌血管生成的影响。方法在缺氧环境下培养人肝癌细胞SK-Hep1,构建HIF1αRNAi慢病毒载体。实验细胞分为阴性对照组(LV-Ctrl)、RNAi组(LV-LV-shHIF1α)、协同作用组(LV-shHIF1α+天冬多糖)。使用CCK8法检测细胞增殖情况、划痕实验和Transwell小室侵袭实验检测细胞迁移侵袭情况、HUVEC诱导的基质胶血管生成实验检测细胞血管生成的能力,免疫印迹法检测相关蛋白表达。结果在缺氧环境下,天冬多糖协同HIF1αRNAi明显抑制肝癌细胞增殖、迁移及血管生成,差异具有统计学意义(P<0.01)。结论天冬多糖协同HIF1αRNAi具有明显的抗肝癌血管生成作用。 Objective To study the effect of asparagus polysaccharide combined with HIF1αRNAi on angiogenesis of liver cancer under hypoxia. Methods Human hepatoma cells SK-Hep1 were cultured under hypoxic environment to construct HIF1αRNAi lentiviral vector. The experimental cells were divided into negative control group(LV-Ctrl), RNAi group(LV-shHIF1α), and synergy group(LV-shHIF1α + asparagine).CCK8 method was used to detect cell proliferation, scratch test and Transwell chamber invasion test to detect cell migration and invasion, HUVEC-induced matrigel angiogenesis test was used to detect cell angiogenesis, and western blot was used to detect related protein expression.Results Under hypoxic environment, aspartic polysaccharides combined with HIF1αRNAi significantly inhibited the proliferation, migration, and angiogenesis of liver cancer cells, and the difference was statistically significant(P <0.01). Conclusion Aspartate polysaccharides and HIF1αRNAi have obvious anti-angiogenic effects in liver cancer.
