摘要
目的探讨预按压和缺血后按压心俞穴对心肌缺血再灌注损伤(Myocardial ischemia-reperfusion injury, MIRI)模型家兔心肌损伤的RISK通路相关激酶的影响。方法将60只新西兰大白兔随机分为正常组、模型组、假手术组、按压心俞组、预按压心俞组,每组12只。结扎冠状动脉左前降支建立心肌缺血再灌注损伤模型;以中医按法刺激器按压心俞穴为干预手段。空白组不予任何干预;模型组、预按压心俞组、按压心俞组给予造模处理;假手术组只做开胸与穿线,不结扎冠脉;按压心俞组在造模成功后给予按压心俞穴操作;预按压心俞组在造模前给予按压心俞穴操作。以高效液相色谱法检测腺苷含量,以Western Blot法和免疫组化检测p-Akt/Akt、p-ERK/ERK、p-PI3K/PI3K。结果预按压心俞组、按压心俞组缺血心肌组织腺苷含量均增高(P<0.05);缺血后按压心俞、预按压心俞均能提高p-PI3K/PI3K、p-Akt/Akt;预按心俞组、按压心俞组有提高p-ERK/ERK比值的趋势。结论预按压心俞和缺血后按压心俞均能减少缺血再灌注损伤,其通过诱发内源性触发物质腺苷来激发RISK通路从而发挥心肌保护效应。
Objective To investigate the effects of pre-pressing and post-ischemic pressing at Xinshu point on RISK pathway-related kinases in rabbits with MIRI(Myocardial ischemia-reperfusion injury). Methods Sixty New Zealand white rabbits were randomly divided into normal group(n=12), model group(n=12), sham operation group(n =12), pressing Xinshu group(n=12) and pre-pressing Xinshu group(n=12). The model of myocardial ischemia-reperfusion injury was established by ligating the left anterior descending branch of coronary artery, and the Xinshu point was pressed with pressing stimulator as intervention. The normal group did not receive any intervention;the model group, the pre-pressing Xinshu group and the pressing Xinyu group were given modeling treatment;the sham operation group only did chest opening and threading, but did not ligate the coronary artery;pressing Xinyu group was given pressing Xinshu point after successful modeling;pre-pressing Xinyu group was given pressing before modeling. The content of adenosine in myocardial tissue of infarcted area was measured by Western Blot method and immunohistochemistry to detect p-Akt/Akt, p-ERK/ERK and pMuPI3 K/PI3 K. Results The content of adenosine in ischemic myocardium increased in pre-pressing Xinshu group and pressing Xinshu group(P=0.001), and pressing Xinshu and pre-pressing Xinshu after ischemia could increase p-PI3 K/PI3 K and p-Akt/Akt(P=0.046 and 0.002, respectively). There was a increased tendency of the pERK/ERK ratio in Pre-pressing Xinyu group and pressing Xinshu group. Conclusion Pre-pressing Xinshu and pressing Xinshu after ischemia reduce ischemia-reperfusion injury, which stimulate RISK pathway by inducing adenosine to play the role of myocardial protection.
