基于网络药理学的苦参黄酮防治糖尿病视网膜病机制研究

摘要 目的:运用网络药理学探讨苦参中黄酮成分防治糖尿病视网膜病的作用机制。方法:通过TCMSP、PharmMapper等数据库检索苦参黄酮成分并预测其作用靶点,根据类药性原则筛选出活性成分;从TTD等数据库检索糖尿病视网膜病的疾病靶点,将药物作用靶点与疾病靶点取交集得到共有靶点;对共有靶点进行蛋白互作网络分析和KEGG通路注释分析,利用Cytoscape软件构建"活性成分-靶点-通路"网络。结果:筛选得到苦参83个活性黄酮成分,涉及DR作用靶点131个。进一步富集分析发现苦参中黄酮成分主要调控AGE-RAGE信号通路、FoxO信号通路、胰岛素抵抗信号通路、肿瘤坏死因子信号通路、PI3K-Akt信号通路、T细胞受体信号通路等166条显著相关通路。结论:网络药理学直观地展示了苦参中黄酮成分多靶点防治糖尿病视网膜病的作用特点,为进一步研究苦参抗糖尿病视网膜病的作用机制提供了科学依据。 Objective To explore the mechanism of flavonoids from Sophora flavescens in the treatment of diabetic retinopathy(DR) by network pharmacology.Methods Firstly, in view of Traditional Chinese Medicine systems pharmacology(TCMSP) and PharmMapper database, the active compounds of flavonoids from Sophora flavescens were screened by Drug-likeness(DL) and the targets of active compounds were predicted. The DR-related targets were collected from Therapeutic Target Database(TTD). Secondly, the shared targets of compounds and DR were imported into the String database to obtain their interaction relationship and combined score for the protein-protein interaction(PPI) network, and perform the GO and KEGG pathway enrichment analyses. Finally, the "compounds-shared targets-pathways" network was constructed for further analysis. Results A total of 83 active ingredients were screened from flavonoids Sophora flavescens, and there were 131 targets involved in DR. Pathway enrichment analysis showed that there were involved in 166 significant metabolic pathways, including AGE-RAGE signaling pathway, Fox O signaling pathway, insulin resistance signaling pathway, tumor necrosis factor signaling pathway, PI3 K-Akt signaling pathway, T cell receptor signaling pathway. Conclusion Network pharmacology demonstrated the characteristics of flavonoids from Sophora flavescens in the treatment of DR with multi-components and multi-targets intuitively, which lay the foundation for further study of the mechanism of flavonoids from Sophora flavescens against DR.
