固本抑瘤Ⅱ号对乳腺癌小鼠RECQL4蛋白和mRNA表达的影响

摘要 目的观察固本抑瘤Ⅱ号及其联合阿霉素对4T1乳腺癌荷瘤小鼠原位瘤生长及RECQL4蛋白和mRNA表达的影响。方法将4T1乳腺癌荷瘤小鼠随机分为对照组、中药组、阿霉素组和中药+阿霉素组,每组6只,每7天测量体重和瘤体积,给药第28天取原位瘤称重,计算抑瘤率,并用蛋白免疫印迹法(Western-blot,WB)和实时定量荧光聚合酶链式扩增反应检测原位瘤组织中RECQL4蛋白和mRNA的表达。结果中药组、阿霉素组、中药+阿霉素组小鼠瘤重均小于对照组(P<0.01),中药+阿霉素组小鼠瘤重均小于中药组、阿霉素组(P<0.01),阿霉素组小鼠瘤重小于中药组(P<0.01)。在给药第28天,中药+阿霉素组小鼠原位瘤体积小于中药组(P<0.05)和对照组(P<0.01),阿霉素组和中药+阿霉素组小鼠体重均较对照组减轻(P<0.01),中药组小鼠体重未见明显影响。与对照组相比,中药组(P<0.05)、阿霉素组(P<0.01)、中药+阿霉素组(P<0.01)中RECQL4蛋白表达和阿霉素组(P<0.05)和中药+阿霉素组(P<0.01)中RECQL4 mRNA表达均偏低。结论固本抑瘤Ⅱ号及其联合阿霉素均能抑制4T1乳腺癌荷瘤小鼠原位瘤的生长,其机制可能与其抑制原位瘤组织中RECQL4蛋白的表达有关。 Objective To observe the effects of Gubenyiliu Ⅱ and its combination with adriamycin on the growth of in-situ tumor and the expression of RECQL4 protein and mRNA in 4 T1 breast cancer tumor-bearing mice. Methods 4 T1 breast cancer tumor-bearing mice were randomly divided into the control group,the Gubenyiliu Ⅱ( GY Ⅱ) group,the adriamycin( ADM) group and the Gubenyiliu Ⅱ and its combination with adriamycin group( GYⅡ + ADM) group,6 mice in each group. Body weight and tumor volume were measured every 7 days. On the 28 th day after administration,the weight of in-situ tumor was measured,the tumor inhibition rate was calculated,and the expression of RECQL4 protein and mRNA in the in-situ tumor tissue were detected by western blot( WB),and the expression of RECQL4 protein and mRNA were detected by using real-time quantitative fluorescent polymerase chain amplification( real-time PCR). Results Compared with the control group,the tumor weight of mice was significantly lower in the GYⅡ group,the ADM group and the GYⅡ+ADM group( P<0.01),the tumor weight of mice in the GYⅡ group was obviously lower than that in the GYⅡ group and the ADM group( P<0.01),and the tumor weight of mice in the ADM group was lower than that in the GYⅡ group( P<0.01). After 28 days administration,the volume of in-situ tumor in the GYⅡ+ADM group was lower than that in the GYⅡ group( P<0.05) and the control group( P< 0. 01). Compared with the control group,the body weight of mice significantly decreased in the ADM group and the GYⅡ + ADM group( P < 0. 01),but there was no significant effect on body weight of mice in the GYⅡ group. Compared with the control group,the expression of RECQL4 protein in the GYⅡ group( P<0.05),the ADM group( P< 0.01) and the GYⅡ+ADM group( P< 0.01) all were decreased. The expression of RECQL4 mRNA in the GYⅡ+ADM group was decreased than that in the control group( P<0.01). Conclusions Both Gubenyiliu Ⅱ and its combination with adriamycin can inhibit the growth of in-situ tumor on breast cancer tumor-bearing mice,its mechanism may be related to the inhibition of the expression of RECQL4 protein in in-situ tumor tissue.
