摘要
本文将猪群中表达频率较高的SLA-1*1301胞外区及SLA-β2 m基因序列片段合成后连接到pET21a(+)原核表达载体上,构建出pET21a-SLA-1*1301和pET21a-SLA-β2m重组表达质粒。通过IPTG诱导大肠杆菌刺激产生大量的相关目标产物,再将所获得的质量比较高的SLA-1*1301胞外区表达蛋白、SLA-β2m蛋白分别与6条软件预测的口蹄疫细胞毒性T淋巴细胞(CTL)表位多肽进行蛋白复性和纯化,通过蛋白纯化仪分析,结果显示,其中4条预测口蹄疫CTL表位多肽可以形成稳定的SLA I-β2m-表位多肽复合体,表明该4条多肽可能为口蹄疫的CTL表位肽,对将来开发细胞免疫疫苗和相关免疫增强剂有重要的指导意义。
In this paper,after synthesizing SLA-1*1301 extracellular region and SLA-β2 m gene sequences with higher expression frequency in pigs,we constructed pET21a-SLA-1*1301 and pET21a-SLA-β2m recombinant plasmids.Large quantities of relevant target proteins were induced in E.coli by IPTG.We got higher quality extracellular proteins of SLA-1*1301 and SLA-β2m,and 6 peptides with foot-and-mouth disease CTL epitopes predicted by software after purification.Protein purification analysis showed that 4 peptides with CTL epitopes formed a stable SLA I-β2m-epitope complex.Our results indicated that 4 peptides were identified to have CTL epitopes of foot-and-mouth disease virus,which will help us develop vaccines in the future.
