摘要
为了探讨运输应激对小鼠脾脏IL-2、IL-4、IL-6、TNF-α和HSP70含量变化的影响,本试验将清洁级小鼠分为对照组(不处理)、处理组1(160 r/min,运输2 h)、处理组2(160 r/min,运输6 h),每组各10只,运输2 h和6 h用恒温培养震荡器模拟运输应激条件进行处理,运用双抗夹心ELISA法测定小鼠脾脏组织IL-2、IL-4、IL-6、HSP-70、TNF-α的浓度。结果表明,与对照组相比,运输2 h后IL-4含量显著降低(P<0.05),运输6 h后含量下降(P=0.083);运输2 h后TNF-α含量有升高趋势(P>0.05),运输6 h后TNF-α含量显著升高(P<0.05)。可见,运输2 h和6 h小鼠脾脏中IL-4水平降低,TNF-α含量升高,提示运输对小鼠免疫功能有影响,脾脏IL-4和TNF-α含量可作为小鼠运输应激的候选评判分子指标。
In order to explore the effects of transportation stress on the changes in the levels of IL-2,IL-4,IL-6,TNF-αand HSP70 in the spleen of mice,we divided clean mice into control group(no treatment)and treatment group 1(160 r/min,transportation for 2 h)and treatment group 2(160 r/min,transportation for 6 h).Each group had 10 animals,and used a constant temperature incubation shaker to simulate transportation stress conditions.The concentration of IL-2,IL-4,IL-6,HSP-70 and TNF-αin mice spleen tissues were measured by double-antibody sandwich ELISA method.The results showed that compared with the control group,the IL-4 content was significantly reduced after 2 h of transportation(P<0.05),and the content decreased after 6 hours of transportation(P=0.083).Compared with the control group,the TNF-αcontent was increased after 2 h of transportation(P>0.05),and the TNF-αcontent was significantly increased after 6 h of transportation(P<0.05).These results indicate that transport has an impact on the immune function of mice,the contents of IL-4 and TNF-αin the spleen can be used as candidate molecular indicators for evaluating transportation stress in mice.
