运输应激对小鼠肾脏显微结构及热休克蛋白表达的影响 被引量:1

摘要 为探究运输应激对小鼠肾脏造成的病理损伤以及热休克蛋白表达的影响,本试验采用H.E.染色、免疫组织化学、蛋白质免疫印迹方法,分析运输条件下小鼠肾脏病变的显微结构,热休克蛋白(HSP27、HSP70和HSP90)的表达部位和表达量。结果显示,运输应激条件下小鼠肾小管和肾间质具有不同程度的病理变化,肾小管病变明显,管腔缩小或消失,上皮细胞发生变性、坏死,细胞排列疏松不规则;肾间质血管有出血、充血现象。免疫组织化学结果表明,HSP27、HSP70和HSP90主要定位于肾小管上皮细胞,但运输应激后HSP90在肾小管基本无表达。免疫印迹分析可知,与对照组相比,应激组HSP27蛋白表达量未见明显变化,HSP70蛋白表达量明显上升(P<0.05),HSP90蛋白表达量明显下降(P<0.05)。表明运输应激会对小鼠肾脏造成一定的损伤,HSP70和HSP90与肾脏的应激损伤存在一定的关联。 In order to explore the effect of transport stress on the pathological damage and heat shock protein expression in mouse kidneys,we used H.E.staining,immunohistochemistry,Western Blot to analyze the microstructure of mouse kidney lesions under transport conditions and heat shock protein(HSP27,HSP70 and HSP90)expression sites and expression levels.The results showed that the renal tubules and renal interstitium of mice under transportation stress had different degrees of pathological changes.The renal tubules were significantly affected.The lumen shrinked or disappearsed,the epithelial cells showed degeneration and necrosis,and the cell arrangement was loose and irregular.Blood vessels had bleeding and congestion.The results of immunohistochemistry showed that HSP27,HSP70 and HSP90 were mainly located in renal tubular epithelial cells,but HSP90 was basically not expressed in renal tubules after transport stress.Western Blot analysis showed that compared with the control group,the expression of HSP27 protein in the stress group did not change significantly,but the expression of HSP70 protein was significantly increased(P<0.05),and the expression of HSP90 protein was significantly decreased(P<0.05).These data suggest that transportation stress can cause certain damage to mouse kidneys,and HSP70 and HSP90 might be related to kidney stress damage.
