金香颗粒预防小鼠大肠杆菌性腹泻的效果观察 被引量:4

摘要 本试验旨在研究金香颗粒在预防小鼠大肠杆菌性腹泻中的作用。同时对空白对照组与感染不给药组灌服生理盐水,对金香颗粒用药组与药物对照组动物进行预防性给药5 d后,除空白对照组外腹腔注射致病性大肠杆菌制造腹泻模型,并观察各组腹泻率与腹泻指数等指标,考察药物对腹泻的预防效果。通过小肠墨水推进率与腹腔液中依文思兰浓度考察药物对肠道蠕动的影响和炎性渗出的抑制作用,通过小肠组织内髓过氧化物酶(MPO)与超氧化物歧化酶(SOD)含量变化情况考察各组炎症程度及抗炎作用。结果显示:与感染不给药组相比,金香颗粒高剂量组的小鼠腹泻指标显著降低(P<0.01);高、中剂量组的小鼠小肠推进率显著降低(P<0.05);金香颗粒高、中、低剂量组均能够显著抑制小鼠腹膜的炎性渗出(P<0.05);高、中剂量组小鼠小肠组织中MPO显著降低(P<0.05),SOD水平显著升高(P<0.01)。试验结果表明,金香颗粒能有效预防小鼠大肠杆菌性腹泻,并且可以通过抑制肠道蠕动、减小炎症引起的渗透性、降低炎症严重程度与减少炎症引起的损伤等作用来预防大肠杆菌性腹泻的发生。 The purpose of this study was to investigate the prophylaxis effect of Jinxiang granule against mouse diarrhea caused by Escherichia coli.Mice were randomly divided into six groups including high,middle and low dose groups of Jinxiang granule,drug control group,infected-nonmedicated group and blank control group.After consecutive prophylactic administration of Jinxiang granules and positive drug for 5 days,the mice except the blank control group were injected intraperitoneally with Escherichia coli.The effects of drugs on intestinal peristalsis and inhibition of inflammatory exudation and the content changes of myeloperoxidase(MPO)and superoxide dismutase(SOD)in small intestine were studied in different groups as the diarrhea evaluation index.The results showed that the diarrhea index in the high dose group was significantly lower than those in the infected-nonmedicated group(P<0.01).In the propulsion experiment of carbon ink,the advancing rates of carbon ink in small intestine in the high and middle dose groups were significantly lower than the infected-nonmedicated group(P<0.05).The inflammatory exudation of mouse peritoneum was significantly inhibited in the high,medium,and low dose groups compared with the infected-nonmedicated group(P<0.05).Compared with the infected-nonmedicated group,the MPO in the mouse small intestine was significantly decreased(P<0.05)and the SOD was significantly increased(P<0.01)in the high and middle dose groups.These results indicate that the Jinxiang granules can prevent the mouse diarrhea caused by Escherichia coli by inhibiting intestinal peristalsis,reducing the permeability caused by inflammation,decreasing the inflammation severity and reducing the injury caused by inflammation.
