急性心肌梗死患者血清微小RNA-143、环氧化酶2表达与心血管事件发生的相关性研究 被引量:4

摘要 目的分析急性心肌梗死(心梗)患者血清微小RNA-143(miR-143)、环氧化酶2(COX-2)的表达情况,并探讨二者与心血管事件发生的相关性。方法选取2018年6月至2019年6月于开封市中医院收治的急性心梗患者93例为心梗组,选择同期我院健康体检者100例为对照组。利用荧光实时定量PCR(qRT-PCR)法检测研究对象血清中miR-143水平,采用酶联免疫吸附法(ELISA)测定血清COX-2水平。采用Pearson法分析miR-143水平与COX-2水平的相关性,分析miR-143水平与COX-2水平与心梗组患者术后6个月心血管事件发生的关系。结果与对照组相比,心梗组患者血清miR-143水平较低,COX-2水平较高(P<0.05)。Pearson相关性分析显示,心梗组患者血清miR-143水平与COX-2水平呈负相关(P<0.05)。miR-143低水平组心血管事件发生率明显高于miR-143高水平组,差异有统计学意义(χ^(2)=4.366,P<0.05)。COX-2高水平组心血管事件发生率明显高于COX-2低水平组,差异有统计学意义(χ^(2)=4.993,P<0.05)。多因素Cox比例风险分析表明,低水平miR-143、高水平COX-2是急性心梗患者心血管事件发生的独立危险因素(HR=1.952,95%CI:1.227~3.106;HR=2.291,95%CI:1.304~4.026,P<0.05)。结论急性心梗患者血清miR-143表达水平下降,COX-2水平上升,且二者与急性心梗患者术后发生不良心血管事件关系密切。 Objective To analyze the expressions of serum miR-143 and cyclooxygenase-2(COX-2)in patients with acute myocardial infarction(AMI),and discuss the correlation among miR-143,COX-2 and major adverse cardiovascular events(MACE).Methods AMI patients were chosen into AMI group(n=93)and healthy controls were chosen into control group(n=100)from Hospital of Traditional Chinese Medicine of Kaifeng City from June 2018 to June 2019.The level of serum miR-143 was detected by using qRT-PCR and level of COX-2 was detected by using ELISA.The correlation between miR-143 and COX-2 was analyzed by using Pearson method,and relationship among miR-143,COX-2 and MACE was analyzed in AMI group after 6 months.Results The level of serum miR-143 was lower and COX-2 level was higher in AMI group than those in control group(P<0.05).The results of Pearson correlation analysis showed that miR-143 and COX-2 had n negative correlation(P<0.05).The incidence rate of MACE was significantly higher in low-miR-143 group than that in high-miR-143 group(χ^(2)=4.366,P<0.05).The incidence rate of MACE was significantly higher in high-COX-2 group than that in low-COX-2 group(χ^(2)=4.993,P<0.05).The results of Cox risk analysis showed that low-level miR-143(HR=1.952,95%CI:1.227~3.106)and high-level COX-2(HR=2.291,95%CI:1.304~4.026)were independent risk factors of MACE in AMI patients(P<0.05).Conclusion The level of serum miR-143 decreases and level of serum COX-2 increases in AMI patient,and they are closely related to MACE incidence.
