缺血性络风内动证大鼠模型构建

摘要 目的依据现代科学技术构建符合临床的、合理的、具有易行性和广泛适用性的缺血性络风内动证动物模型。方法将70只体重为(260±20)g清洁级SD雄性大鼠随机分成模型组40只和假手术组30只,模型组开胸后将月桂酸钠(0.75 mg/ml)和同种异体血栓微颗粒(2.5 mg)的混合液0.2 ml注入其主动脉根部,假手术组注入等体积纯水,两组分别于术后1 h、术后3 h、术后1 d、术后3 d、术后1 w时取材,观察其一般情况、病理组织以及肌钙蛋白I(cTnI)和血管假血友病因子(vWF)变化。结果各组实验动物起点和终点体重之间差异无统计学意义,整体死亡率为20%,成模率为80%;组织病理学变化:模型组术后1 h、3 h以微血栓表现为主,3 h后出现血管内皮损伤及炎细胞浸润,术后3 d及术后1周可见多发性微梗死灶,假手术组未见明显血栓或梗死灶。模型组各时间点cTnI均高于同时间点假手术组;模型组vWF术后1 h至3 d浓度均低于假手术组,术后1周时高于假手术组。结论通过向大鼠主动脉根部注入同种异体血栓微颗粒(2.5 mg)和月桂酸钠(0.75 mg/ml)的混合液(0.2 ml),可初步建立缺血性络风内动证动物模型。 Objective To establish a clinical,reasonable,feasible and widely applicable animal model of pattern of internal stirring of ischemic collateral wind based on modern science and technology.Methods Male SD rats(weight=260±20 g,n=70)were divided randomly into model group(n=40)and sham-operation group(n=30).The model group was given injection of mixed solution(0.2 ml)of sodium laurate(0.75 mg/ml)and allograft thrombosis microparticles(2.5 mg)into the aortic root and sham-operation group was given equal-volume of pure water after thoracotomy.The blood samples and myocardial samples were collected for observing the changes of general state,pathological tissue,cardiac troponin I(cTnI)and von Willebrand factor(vWF)in 2 groups after the operation respectively for 1h,3h,1d,3d and 1 week.Results The difference in beginning weight and ending weight had no statistical significance between 2 groups.The overall mortality was 20%and modeling rate was 80%.The histopathological changes included microthrombus manifestations in model group after operation for 1h and 3.The vascular endothelial injury and inflammatory cell infiltration were observed after operation for 3 h,and multiple micro-infarctions was observed after operation for 3d and 1 week in model group.There were no significant thrombosis and infarction observed in sham-operation group.The level of cTnI was higher in model group than that in sham-operation group at all time points.The level of vWF was lower in model group than that in sham-operation group after operation from 1h to 3d,and higher in model group than that in sham-operation group after operation for 1 week.Conclusion The animal model of pattern of internal stirring of ischemic collateral wind can be initially established through the injection of mixed solution(0.2 ml)of sodium laurate(0.75 mg/ml)and allograft thrombosis microparticles(2.5 mg)into rat aortic root.
