基于生物信息学分析LOXL2在肺腺癌中的表达及临床意义 被引量:1

摘要 目的基于多数据库探讨LOXL2在肺腺癌中的表达水平差异和临床意义。方法利用UALCAN分析平台对LOXL2基因在TCGA数据库中的m RNA表达量进行泛癌分析;利用Oncomine和GEPIA数据库对LOXL2基因在肺腺癌中的表达进行荟萃分析;使用K-Mplot对肺腺癌中LOXL2表达量与预后关系进行分析;利用UALCAN分析LOXL2基因表达水平与临床特征的关系;使用MEXPRESS数据库对LOXL2启动子区域甲基化水平进行分析;使用String数据库构建相关蛋白网络并进行功能富集分析。结果多个数据库分析显示LOXL2在肺腺癌组织中表达水平高于正常组织(P<0.05),且与TNM分型相关;高表达LOXL2的肺腺癌患者总生存率和首次进展时间低于LOXL2低表达患者(P<0.05)UALCAN与MEXPRESS数据库分析表明,LOXL2基因启动子区域甲基化水平在肿瘤组织中较高。String数据库分析显示,MFAP5、MFAP2、FBLN5、PCOLCE、ELN、LOX、LOX1、LOX3、BMP1、TLL1等与LOXL2关系密切。结论通过挖掘并分析肿瘤基因数据库信息表明,LOXL2在肺腺癌组织中呈高表达,且与患者预后不良相关,为后续深入研究LOXL2在肺腺癌进行过程中的作用提供重要理论依据。 Objective To explore the expression pattern and clinical significance of LOXL2 in lung adenocarcinoma based on multi-database analysis.Methods UALCAN was used to analyze the mRNA expression of LOXL2 in TCGA database for pan-cancer analysis.LOXL2 gene expression in lung adenocarcinoma was analyzed using Oncomine and GEPIA database.K-Mplot was used to analyze the relationship between LOXL2 expression and prognosis in lung adenocarcinoma.The relationship between LOXL2 expression and HCC clinical characteristics was evaluated using UALCAN;DNA methylation in LOXL2 gene was assessed using online MEXPRESS tool and protein-protein interaction networks and functional enrichment analysis were analyzed using String database.Results The expression level of LOXL2 in lung adenocarcinoma tissues was higher than that in normal tissues based on multiple databases(P<0.05),and was related to TNM classification(P<0.05).The os and PPS of patients with high expression of LOXL2 were significandly lower than those with low expression of LOXL2(P<0.05).Aanalysis of UALCAN database and MEXPRESS database showed that the methylation level of LOXL2 gene promoter region is higher in tumor tissues.Analysis in the String database showed that MFAP5,MFAP2,FBLN5,PCOLCE,ELN,LOX,LOX1,LOX3,BMP1 and TLL1 are closely related to LOXL2.Conclusion By exploring and analyzing information from the tumor gene database,it was shown that LOXL2 was strongly expressed in the tissues of lung adenocarcinoma and was associated with poor patient prognosis,providing an important theoretical basis for an in-depth study of the role of LOXL2 in the progression of lung adenocarcinoma.
