基于网络药理学探讨清肠化湿方治疗溃疡性结肠炎的作用机制 被引量:3

摘要 目的:运用网络药理学方法探讨清肠化湿方治疗溃疡性结肠炎(ulcerative colitis,UC)的作用靶点与信号通路,从而解释其治疗湿热型UC的分子作用机制。方法:应用中药系统药理学技术平台(TCMSP)筛选清肠化湿方的潜在活性成分(OB≥30%,DL≥0.18)及作用靶点。利用Cytoscape软件,通过TCMSP、GeneCards数据库构建“潜在活性成分-作用靶点-疾病”网络。运用STRING平台,构建蛋白-蛋白相互作用网络模型,找出核心靶点。通过R软件对潜在靶点进行GO、KEGG富集分析。结果:网络分析结果表明,清肠化湿方中排名前4位的活性成分是:槲皮素、山奈酚、汉黄芩素、黄芩素。从清肠化湿方中找到210个基因靶点,UC中有4213个靶点,清肠化湿方治疗UC的关键靶点包括AKT1、IL-6、VEGFA、CASP3等,GO富集分析显示这些基因靶点主要影响核受体、类固醇激素受体、细胞因子受体结合、细胞因子活性。KEGG富集分析表明,AGE-RAGE信号通路、IL-17信号通路、TNF等通路富集较多。结论:本研究阐述了清肠化湿方治疗UC的物质基础及作用机制,为未来研究提供了理论基础和研究方向。 Objective:To explore the target and signal pathway of Qingchang Huashi decoction(QCHSD)in the treatment of ulcerative colitis(UC)by using network pharmacology and to explain its molecular mechanism of action in the treatment of UC of damp heat.Methods:TCMSP was used to screen the potential active components(OB≥30%,DL≥0.18)and targets of QCHSD.The network of"potential active ingredients-target-disease"was constructed by using the database of TCMSP and GeneCards.The protein protein interaction(PPI)network model was constructed by the String platform to find core targets.GO and KEGG enrichments of potential targets were analyzed by R software.Results:The results of network analysis showed that quercetin,kaempferol,wogonin,and baicalein were the top four active ingredients in QCHSD.210 gene targets were found in QCHSD,4213 in UC.The key targets of QCHSD in treating UC included AKT1,IL-6,VEGFA,CASP3,etc.GO enrichment analysis showed that these gene targets mainly affected nuclear receptor,steroid receptor,cytokine receptor binding,cytokine activity,etc.KEGG enrichment analysis showed that AGE-RAGE signaling pathway,IL-17 signaling pathway,and TNF were more abundant.Conclusion:This study illustrates the material basis and mechanism of QCHSD in the treatment of UC,which provides a theoretical basis and research direction for future research.
