复方黄芪饲料添加剂高效液相指纹图谱研究 被引量:2

摘要 试验旨在更好地控制复方黄芪饲料添加剂的质量品质,建立复方黄芪饲料添加剂(compound Astragalus feed additive,CAFA)指纹图谱分析方法。采用高效液相色谱法(HPLC)建立不同批次饲料添加剂指纹图谱,以乙腈-0.15%甲酸水溶液梯度洗脱,在254 nm下进行检测。对研究结果进行相似度分析,并结合化学模式识别技术聚类分析(CA)与主成分分析(PCA)进行质量研究。结果显示,CAFA中主要含黄酮和异黄酮类化学成分,指纹图谱测定出31个共有峰,10批样品相似度均大于0.9。对照品指认出毛蕊异黄酮葡萄糖苷、芒柄花黄素和左旋紫草素。CA和PCA结果表明,由于产地不同,样品之间质量有所差异。研究表明,HPLC法指纹图谱可应用于不同批次CAFA的质量评价。 The study aimed to establish a fingerprint analysis method of compound Astragalus feed additive(CAFA)for control the quality of compound astragalus feed additives.High performance liquid chromatography(HPLC)was used to establish fingerprints of different batches of CAFA,and the detection was carried out at 254 nm with a gradient of acetonitrile-0.15%formic acid aqueous solution.The similarity analysis of the research results were combined with chemical pattern recognition technology cluster analysis(CA)and principal component analysis(PCA)for quality assessment.The results showed that CAFA mainly contains flavonoids and isoflavones.According to the fingerprint of CAFA,31 common peaks were determined,the similarity of ten batches of samples were all greater than 0.9.The reference refered to verbasil glucoside,formononetin and shikonin.CA and PCA showed differences in quality between samples due to different origins.The results indicated that fingerprint of HPLC method can accurately provide quality assurance in the CAFA.
