摘要
目的本文旨在介绍一种有效培养出大鼠的原代血管平滑肌细胞的方法。方法选取12只10周龄的自发性高血压大鼠采用腹腔注射维生素D360万/U后联合高脂饲料喂养3周,对照组选12只10周龄的普通大鼠喂养普通饲料3周;然后取材大鼠胸主动脉后HE染色。采用组织贴壁法培养大鼠胸主动脉血管平滑肌细胞,并用α-SMA抗体采用细胞免疫荧光法来鉴定。结论动脉粥样硬化形成的自发性高血压大鼠更容易培养出纯度更高的原代血管平滑肌细胞。
Objective The article aims to introduce a method for the effective culture of primary vascular smooth muscle cells in rats.Methods Twelve spontaneous hypertensive rats aged 10 weeks were raised up with high fat diet for 3 weeks after intraperitoneally injecting 600,000/U of vitamin D3.In the control group,12 normal rats aged 10 weeks were fed with normal feed for 3 weeks.the thoracic aorta of the rats will be removed and observed with HE staining.The vascular smooth muscle cells will be cultured with tissue adherent method and α-SMA antibody was evaluated by immunofluorescence method.Conclusion The primary vascular smooth muscle cells with higher purity were more likely to be cultured in the spontaneously hypertensive rats with atherosclerosis.
