油莎豆CebZIP1基因的克隆及表达分析 被引量:3

摘要 bZIP蛋白是参与植物生长发育和抗性调控的一类转录因子。为探究bZIP在能源植物油莎豆中的生物学功能,从盐胁迫的转录组数据中筛选到1个bZIP家族基因,命名为CebZIP1,随后对CebZIP1基因进行克隆、生物信息学分析和基因表达模式分析。结果表明,CebZIP1基因的开放阅读框(ORF)全长为948 bp,共编码315个氨基酸,蛋白质分子质量为34.13 ku,理论等电点为5.02,经预测定位于细胞核内;二级结构以α-螺旋和无规则卷曲为主。系统发育分析表明,油莎豆与禾本科亲缘关系较近。组织特异性表达模式显示,CebZIP1在所有组织中均能检测到表达,表达量由高到低依次为叶片、根和块茎,揭示CebZIP1基因可能参与调控油莎豆的生长发育过程。同时,CebZIP1基因的表达水平受干旱、高盐及低温胁迫诱导,推测其可能在油莎豆非生物胁迫过程中发挥重要功能。研究结果可为更深入揭开bZIP转录因子在油莎豆发育及非生物胁迫应答中的功能提供一定理论依据。 bZIP proteins are transcription factors involved role in plant growth,development and resistance regulation.To explore the biological function of bZIP in Cyperus esculentus,a bZIP family gene was identified based on the RNA-Seq data,then the CebZIP1 gene was cloned and analyzed by bioinformation tools.The gene expression pattern was analyzed by RT-qPCR.The results showed that the length of CebZIP1 open reading frame(ORF)was 948 bp,encoded 315 amino acid,which was located in the nucleus.The relative molecular quality of the protein was 34.13 ku,and the theoretical isoelectric point was 5.02.The secondary structure was mainly composed ofα-helix and random coil.The phylogenetic analysis showed that Cyperus esculentus was closely related to Gramineae.The RT-qPCR results showed that the expression pattern of CebZIP1 gene was found to be high expression in leaf,followed by root and low expression in tuber,suggesting that CebZIP1 gene might be involved in the regulation of the growth and development of Cyperus esculentus.In addition,the expression level of CebZIP1 gene was induced by drought,high salt and low temperature,which suggested that CebZIP1 might play an important role in the abiotic stress processes in Cyperus esculentus.This study result will provide a basis for further research on the function of bZIP transcription factor in the development and abiotic stress response of Cyperus esculentus.
