摘要
目的探讨一种新型的采用微创技术获取脂肪来制备脂肪来源干细胞(adipose derived stem cells,ADSCs)的方法。方法自2018年5月至2019年8月在北京雷华医疗美容诊所通过腹部穿刺方法获取5例患者腹部脂肪(0.5~1.5 g)组织进行分离培养,扩增ADSCs,并进行干细胞的分化与鉴定分析。结果经过对ADSCs的增殖能力、特异性表面标志物的流式鉴定,成脂分化、成骨分化、软骨分化、血管内皮细胞分化及神经分化的多项分化能力的鉴定,确定了分离出的细胞为脂肪间充质干细胞。结论采用微创技术可以分离获取ADSCs。
Objective To explore a new minimally invasive method for isolation of adipose-derived stem cells(ADSCs).Methods From May 2018 to August 2019,0.5 g to 1.5 g abdominal adipose tissues in 5 patients were obtained by puncture needles.After isolation and culture,the differentiation,identification and analysis of ADSCs were conducted.Results After identification of proliferation of ADSCs,flow cytometry analysis,adipocyte differentiation,osteogenic differentiation,chondrogenicd ifferentiation,vascular endothelial cells and neuronal differentiation,the isolated cells were confirmed asadipose mesenchymal stem cells.Conclusion The ADSCs can be isolated and obtained by minimally invasive technology.
