温和灸对大鼠肌筋膜激痛点肌卫星细胞激活及成肌修复的影响

摘要 目的:探讨在SD大鼠肌筋膜激痛点(myofascial trigger point,MTrP)模型上,温和灸对骨骼肌干细胞-肌卫星细胞(skeletal muscle satellite cell,SMSC)激活和成肌修复的影响。方法:30只SD大鼠随机分为正常组、模型组、温和灸组给予相应处理,并在造模开始的第30天和第60天取MTrP处或相同部位标本,HE染色检测MTrP部位的组织修复情况,免疫荧光法检测SMSC标志Pax7的变化。结果:第30天和第60天正常组大鼠肌细胞呈规则多边形、间隙小、未见明显炎性细胞浸润;模型组肌细胞呈不规则形和类圆形并萎缩、大小不等形成收缩结节且炎性细胞浸润;温和灸组肌细胞趋向规则,紊乱程度和炎性细胞浸润较模型组明显改善。正常组、模型组、温和灸组3组中肌细胞面积百分比在第30天分别为(70.73±7.85、18.63±2.98和44.09±5.23%),第60天分别为(74.98±7.33、12.06±3.24和53.58±6.06%),均为正常组>温和灸组>模型组(F=104.14和152.11,P均<0.05)。3组中Pax7荧光强度在第30天分别为(211.30±18.06、42.75±12.25和138.19±25.95(μm^(2)/mm^(2)×10^(3))),第60天分别为(199.80±28.06、27.34±3.11和187.49±37.46(μm^(2)/mm^(2)×10^(3))),均为正常组>温和灸组>模型组(F=93.24和63.10,P均<0.05)。结论:SD大鼠MTrP温和灸可减轻炎症,避免SMSC过度激活,维持SMSC的更新。这可能是温和灸促进MTrP成肌修复的作用机制之一。 Objective:To investigate the effects of mild moxibustion on activation of skeletal muscle stem cell-skeletal muscle satellite cell(SMSC)and myogenesis repair in SD rat model of myofascial trigger point(MTrP).Methods:Thirty SD rats were randomly divided into control group,modeling group and group of mild moxibustion intervention.Rats were harvested at the 30th and 60th day after dealing with correspondingly.HE staining detected the repair of MTrP,and Pax7 was detected by immunofluorescence for evaluation of SMSC.Results:The skeletal muscle cells showed regular polygons with small gaps,and no obvious inflammatory cell infiltration in control group.However,muscle cells in modeling group showed irregular shapes and rounded shapes,with atrophy and different sizes,contraction knots and infiltration of inflammatory cells.Muscle cells in group of mild moxibustion intervention tended to be regular with less disturbance and inflammatory cells infiltration compared with that of the modeling group.The percentage of muscle cell area in the three groups was(70.73±7.85,18.63±2.98)and(44.09±5.23%)respectively on the(30th day,and 74.98±7.33,12.06±3.24 and 53.58±6.06%)respectively on the 60th day,which meant that percentage in modeling group was the least(F=104.14 and 152.11,P<0.05).Pax7 fluorescence intensity in the 3 groups was(211.30±18.06,42.75±12.25),and(138.19±25.95(μm^(2)/mm^(2)×10^(3))respectively on the 30th day,and(199.80±28.06,27.34±3.11,and 187.49±37.46(μm^(2)/mm^(2)×10^(3))respectively on the 30th day,which meant that fluorescence intensity in modeling group was the least(F=93.24 and 63.10,P<0.05).Conclusion:Mild moxibustion on MTrP of SD rats can inhibit inflammation,maintain the renewal of SMSC out of depletion.This may be one mechanism for mild moxibustion to promote skeletal muscle repair in MTrP.
