无创法诱发Wistar大鼠子宫腺肌病模型的建立 被引量:2

摘要 目的:比较无创法和开腹法建立子宫腺肌病动物模型的成模效果,评价通过无创法经垂体移植术建立子宫腺肌病动物模型的可行性和有效性。方法:120只雌性Wistar大鼠随机分为无创造模组和开腹造模组,每组60只,另有10只大鼠作为空白对照组。无创造模组大鼠利用本课题组自制专利经阴道-宫颈途径将雄鼠垂体植入雌鼠宫腔;开腹造模组大鼠开腹后进行宫腔操作将雄鼠垂体放入子宫内;空白对照组不予任何处理。3个月后以子宫形态学观察、子宫HE染色评分及泌乳素(PRL)、发病率等指标评价成模效果。结果:3个月后,无创组和开腹组子宫HE染色评分、PRL均显著高于空白对照组(P<0.01),两造模组间各项指标比较差别无统计学意义。结论:无创法可成功诱发大鼠子宫腺肌病,造模的各项评价指标与开腹法无显著差异,成功率高且操作更加简单易行,可作为子宫腺肌病动物模型造模方式的优选。 Objective:The effects of noninvasive and open abdominal methods for establishing animal models of adenomyosis were compared,and the feasibility and effectiveness of establishing animal models of adenomyosis by non-invasive method through pituitary transplantation were evaluated.Methods:A total of 120 female Wistar rats were randomly divided into non-invasive group and open group,60 rats in each group,and 10 rats were used as blank control group.In the non-invasive group,the male pituitary was implanted into the uterus of the female by the‘vaginal-cervical’route.The rats in the open group were treated with uterine cavity after the laparotomy.The blank control group was not given any treatment.Three months later,the model was evaluated by uterine morphology,uterine HE staining score,prolactin(PRL),and incidence.Results:After 3 months,the HE staining score and PRL in the non-invasive group and the open group were significantly higher than those in the blank control group(P<0.01),but there was no significant difference between the two groups.Conclusion:The noninvasive method can successfully induce adenomyosis in rats.There is no significant difference between the evaluation indexes of modeling and the open group.The success rate is high and the operation is simpler and easier.It is the best model for adenomyosis animal model.
