干旱环境中健康大鼠外周血CD_(3)^(+)、CD_(4)^(+)、CD_(8)^(+)及CD_(4)^(+)/CD_(8)^(+)值的变化 被引量:8

Effects of drought environment on expression of CD_(3)^(+), CD_(4)^(+), CD_(8)^(+) and CD_(4)^(+)/CD_(8)^(+) ratio in peripheral blood of healthy rats

摘要 目的:基于干旱环境对大鼠外周血CD_(3)^(+)、CD_(4)^(+)、CD_(8)^(+)及CD_(4)^(+)/CD_(8)^(+)值的变化探讨燥邪的致病性。方法:60只SPF级SD大鼠按体质量随机分为正常对照24d(N-24d)组、正常对照48d(N-48d)组、干燥24d(D-24d)组、干燥48d(D-48d)组、干旱风沙24d(D&D-24d)组、干旱风沙48d(D&D-48d)组,并分别普通饲料喂养在常温常湿环境、干燥环境和干旱风沙环境中,在实验干预的第25天经眼内眦静脉采集N-24d组、D-24d组和D&D-24d组大鼠外周血,余组在第49天同法采血,并随即进行流式细胞学检测。结果:D-48d组外周血CD_(8)^(+)表达低于D-24d组(P<0.05),D-48d组CD_(4)^(+)/CD_(8)^(+)值高于D-24d组(P<0.05);D&D-24d组和D&D-48d组外周血CD_(3)^(+)表达均低于N-48d组(P<0.05),D&D-24d组外周血CD_(8)^(+)表达分别低于N-48d组和D&D-48d组(P<0.05),D&D-24d组外周血CD_(4)^(+)表达、CD_(4)^(+)/CD_(8)^(+)值均高于N-48d组和D&D-48d组(P<0.05)。结论:免疫机能亢进可能是燥邪致病的微观机制之一,外周血CD_(3)^(+)、CD_(4)^(+)、CD_(8)^(+)及CD_(4)^(+)/CD_(8)^(+)值可做为西北燥证证候机制研究的观察指标之一。 Objective:To explore mechanisms of‘pathological dry syndrome causing by dryness evil’and to verify the pathogenicity of dryness evil based on expression of CD_(3)^(+),CD_(4)^(+),CD_(8)^(+)and CD_(4)^(+)/CD_(8)^(+)ratio in peripheral blood of healthy rats.Methods:Sixty SPF grade SD rats were randomly divided into normal control 24 d group(N-24 d group),normal control 48 d group(N-48 d group),dry 24 d group(D-24 d group),dry 48 d group(D-48 d group),dry wind sand 24 d group(D&D-24 d group),dry wind sand 48 d group(D&D-48 d group)according to body weight,and were fed with common food respectively in normal temperature and humidity environment,dry environment,and arid sandstorm environment,then to collect peripheral blood of rats in N-24 d group,D-24 d group,D&D-24 group through medial canthus vein on the 25 th intervention day,so do the other rats on the 49 th intervention day,still to put peripheral blood detected by flow cytometry skill immediately.Results:CD_(8)^(+)expression in peripheral blood of D-48 d group was lower than it in D-24 d group respectively(P<0.05),CD_(4)^(+)/CD_(8)^(+)ratio of D-48 d group was higher than D-24 d group(P<0.05).CD_(3)^(+)expression in peripheral blood of D&D-24 d group and D&D-48 d group were lower than it in N-48 d group(P<0.05).CD_(8)^(+)expression in peripheral blood of D&D-24 d group was lower than N-48 d group and D&D-48 d group,respectively(P<0.05).CD_(4)^(+)expression in peripheral blood and CD_(4)^(+)/CD_(8)^(+)ratio of D&D-24 group were higher than N-48 d group and D&D-48 d group,respectively(P<0.05).Conclusion:Immune hyperfunction may be one of the micro mechanisms of pathogenicity of dryness evil.Expression of CD_(3)^(+),CD_(4)^(+),CD_(8)^(+)and CD_(4)^(+)/CD_(8)^(+)ratio in peripheral blood can be the observational index when we research the micro mechanisms of northwest dryness syndrome.
