摘要
目的:探讨利用单细胞RNA测序技术研究膀胱癌患者外周血特征的效果,为研究膀胱癌患者外周血提供新的策略。方法:分别收集膀胱癌患者和健康人的外周血,提取外周血单个核细胞(PBMC)进行单细胞RNA测序。对测序结果进行差异基因分析,根据基因表达情况降维聚类分群,定义各细胞亚群的细胞类型和比较各亚群之间的差异。结果:单细胞RNA测序检测到健康人PBMC有7523个细胞,膀胱癌患者PBMC有5289个细胞,根据上述细胞的基因的表达情况将细胞聚类分为15个亚群。健康人和膀胱癌患者的PBMC细胞类型具有相似性但细胞亚型的分布具有差异性,尤其是CD8^(+)和CD4^(+)T细胞,并且在膀胱癌患者PBMC中发现了膀胱癌患者特有的细胞亚群。结论:单细胞RNA测序能够实现对PBMC做到精细分群,通过展示来自健康人和膀胱癌患者PBMC的表达谱,能够更精准的反映PBMC细胞间的差异,为研究膀胱癌患者PBMC提供了一个新的研究方案。
Objective:To explore the effect of studying the characteristics of peripheral blood in patients with bladder cancer by using single-cell RNA sequencing to,and to provide a new strategy for the study of peripheral blood in bladder cancer patients.Method:Peripheral blood was collected from bladder cancer patient andhealthy individuals,and then PBMCs were extracted for single-cell RNA sequencing.The results of sequencing were analyzed to identify the differential genes,The results of sequencing were analyzed to identify the differential genes.According to the gene expression,the cells were clustered after dimensionreduction,then we define the cell types of each cell subgroup and compare the differences among them.Results:Single-cell RNA sequencing detected 7,523 cells from healthy individuals and 5,289 cells from bladder cancer patient,and the cells were clustered into 15 subpopulations based on the gene expression.The cell types of PBMCs from healthy individuals and tumor patient had similarity,but the distribution of cell subtypes was different,especially the CD8^(+)and CD4^(+)T cells,and a subtype was specifically identified in PBMC from bladder cancer patient.Conclusion:Single-cell RNA sequencing enables fine clustering of PBMC,and by demonstrating the expression profiles of PBMC from healthy donors and bladder cancer patient,it can more accurately reflect the differences between PBMCs,and provide a new protocol for studying PBMCs in bladder cancer patients.
