摘要
目的:探讨不同浓度和接收速度对聚己内酯(PCL)、聚乳酸(PLA)和聚乙烯醇(PVA)电纺纳米纤维的纤维结构、直径、机械性能以及细胞活力的影响。方法:利用静电纺丝技术对不同浓度的PCL、PLA和PVA溶液进行电纺,同时设定不同的接受速度;通过扫描电镜(SEM)和力学仪器对PCL、PLA和PVA纳米纤维膜的平均直径和机械性能(拉伸应力、拉伸应变和杨氏模量)进行检测,并观察纤维形态。将SD乳鼠的骨髓间充质干细胞种植于PCL、PLA和PVA纳米纤维膜上,通过CCK-8和活死细胞染色检测细胞增值能力。结果:当接收速度为20 r/min时,3种电纺纤维的直径大小和细胞活力随着浓度的增加而增大(均P<0.05);当浓度相同时,3种电纺纤维的直径随着接收速度的增加而减小,同时,细胞活力随着接收速度的增加而增加(均P<0.05)。随着浓度的增加,PCL、PVA和PLA 3种纤维膜的力学性能先增加后降低,不同的接收速度对3种纤维膜的机械性能均有影响(P<0.05)。结论:PCL、PLA和PVA电纺纤维的形态结构、直径、机械性能和细胞活性受到电纺溶液浓度和接收速度影响。
Objective:To investigate the effect of different concentrations and receiving speeds on the fiber structure,diameter,mechanical properties and the cell vitality of polycaprolactone(PCL),polylactic acid(PLA)and polyvinyl alcohol(PVA)electrospinning nanofibers.Methods:PCL,PLA and PVA nanofiber membranes were carried out by electrospinning technology with different concentration solutions or different receiving speeds.The morphology,diameter distribution and mechanical properties of PCL,PLA and PVA nanofiber membranes were measured by scanning electron microscope(SEM)and mechanical instrument.The bone marrow mesenchyml stem cells(BMSCs)from SD rats were inoculated on PCL,PLA and PVA nanofiber membranes,and the proliferation of cells was tested by CCK-8 and Calcein-AM/PI.Results:The diameters of the electrospinning fibers were increased with higher concentration at 20 r/min receiving speed,and were decreased with faster receiving speed.The cell vitality were increased with higher concentration at 20 r/min receiving speed,and were increased with faster receiving speed.The mechanical properties of electrospinning fibers were improved with the increase of the concentration,and decreased with the increase of receiving speed.Conclusion:Electrospinning solution concentration and receiving speed had influence on diameter distribution,mechanical properties and thecell vitality of PCL,PLA and PVA nanofiber membranes.
