分子马达相关蛋白在小鼠神经系统中表达和分布

摘要 目的探讨沿微管运动的驱动蛋白(kinesin)、动力蛋白(dynein)和动力蛋白激活蛋白(dynactin)在小鼠神经系统中的表达和分布。方法以1.5月龄野生型C57BL/6 J小鼠眼、脑、脊髓、坐骨神经和肌肉组织为材料,利用蛋白质印迹法(Western blot)检测不同部位中分子马达相关蛋白表达分布,并进一步研究脊髓和坐骨神经中蛋白质含量差异。结果Dynein亚基DIC(dynein intermediate chain)、DLIC(dynein light intermediate chain),Dynactin亚基DCTN4、DCTN5以及Kinesin在小鼠眼、脑、脊髓、坐骨神经和肌肉中均有分布,但表达水平不同。与脊髓相比,小鼠坐骨神经中DIC、DLIC、DCTN4、DCTN5、Kinesin heavy chain含量均显著下降(P<0.05)。结论小鼠神经系统多区域均分布有分子马达相关蛋白,但与脊髓(胞体)相比,坐骨神经(轴突束)中分子马达相关蛋白的含量相对较少。而分子马达相关蛋白是调控轴突运输关键因子,因此其含量或功能改变可能更易引起轴突损伤。 Objective To investigate the expression and distribution of motor proteins which moving along microtubule in the nervous system of mice.Methods The motor protein Kinesin,Dynein and Dynactin distribution in the eye,olfactory bulb,cortex,hippocampus,striatum,midbrain,cerebellum,brainstem,spinal cord,sciatic nerve and muscle,and the expression of motor proteins in spinal cord and sciatic nerve of 1.5 months wild-type C57 BL/6 J mouse were examined by western blotting.Results DIC,DLIC,DCTN4,DCTN5,Kinesin heavy chain had different expression levels in eye,brain,spinal cord,sciatic nerve and muscle.Compared to spinal cord,the sciatic nerve showed decreased level of DIC,DLIC,DCTN4,DCTN5,Kinesin heavy chain(P<0.05).Conclusions Motor proteins were found in the nervous system of mice.However,compared with the spinal cord(cell body),the content of motor proteins in sciatic nerve(axonal tract)was relatively low.And motor proteins is a key factor in the regulation of axon transport,the content or function changes of motor proteins may be more likely to cause axon injury.
