食管鳞癌细胞KYSE150外泌体对细胞恶性表型的影响 被引量:1

摘要 目的探讨食管鳞癌细胞KYSE150外泌体对细胞增殖、侵袭迁移的影响。方法培养KYSE150并用试剂盒法提取外泌体,采用透射电镜、纳米粒子示踪分析、蛋白免疫印迹法、吞噬实验等对外泌体进行鉴定。将外泌体与KYSE150共培养,采用细胞划痕实验比较外泌体对细胞增殖能力的影响;采用Transwell实验比较外泌体对细胞迁移能力的影响。结果提取后的外泌体经过纳米粒子示踪分析提示颗粒大小30~200 nm,电镜形态正常,免疫印迹实验显示CD9和CD81均有条带出现,且外泌体可被自身吞噬。与PBS对照组的细胞迁移率(47.67±2.52)%和迁移细胞数(147.33±5.89)个比较,外泌体实验组的细胞迁移率(64.67±3.05)%和迁移细胞数(233.22±4.58)个明显增高,差异有统计学意义(P<0.05),提示外泌体实验组的细胞增殖与侵袭迁移能力增强。结论外泌体能显著促进食管鳞癌细胞运动能力。 Objective To investigate the effects of exosomes from esophageal squamous cell.Methods Exosomes were extracted by exosome extraction kit,and identified by electromicroscope,nanoparticle tracer analysis(NTA),western blot and phagocytosis test.The effects of KYSE150 exosomes on proliferation were evaluated by cell scratch test and the migration ability were estimated by transwell test.Results NTA analysis showed that the size of the extracted exosomes was 30-200 nm,and the morphology of the exosomes was normal under electron microscope.Western blotting showed that both CD9 and CD81 had bands,and the exocrine body could be swallowed by itself.Compared with PBS Control Group(47.67±2.52)%and(147.33±5.89),the cell migration rate(64.67±3.05)%and the cell migration number(233.22±4.58)in exocrine body experiment group were significantly higher(P<0.05),these results suggest that the ability of cell proliferation,invasion and migration in exocrine body experimental group is enhanced.Conclusion Exosomes can promote the proliferation,invasion and migration of ESCC cells significantly.
