miR-155与AGTR1在子痫前期患者胎盘组织中的表达 被引量:3

摘要 目的分析微小RNA155(miR-155)与血管紧张素Ⅱ受体1(AGTR1)在子痫前期患者胎盘中表达的相关性。方法收集2019年11月~2020年7月唐山市妇幼保健院及唐山市工人医院行剖宫产术分娩的29例正常孕产妇(对照组)、15例子痫前期孕产妇(子痫前期组)及21例重度子痫前期孕产妇(重度子痫前期组)的胎盘组织,应用实时荧光定量PCR、Western blotting技术检测miR-155与AGTR1的表达。结果miR-155在对照组、子痫前期组及重度子痫前期组中的表达量分别为(1.18±0.12)、(1.57±0.07)及(1.98±0.10),呈升高趋势,各组间差异有统计学意义(F=384.2,P<0.001);AGTR1蛋白在对照组、子痫前期组及重度子痫前期组中的表达量分别为(0.86±0.08)、(0.57±0.04)及(0.29±0.07),呈下降趋势,各组间差异有统计学意义(F=99.4,P<0.001)。结论在病情程度不同的子痫前期患者胎盘组织中,miR-155与AGTR1的表达情况不同;随着疾病的加重,miR-155表达升高,AGTR1表达降低;推测miR-155可能通过调控AGTR1影响子痫前期的发病。 Objective To make a correlation analysis of expressions of miR-155 and AGTR1 in placental tissues of preeclampsia(PE)patients.Methods A total of 65 pregnant women were gathered in Tangshan Maternal and Child Health Care Hospital and Tangshan Gongren Hospital from November 2019 to July 2020,their placental tissues were collected and divided into normal pregnant women(control group,n=29),PE patients(PE group,n=15)and severe PE patients(severe PE group,n=21).Real-time quantitative PCR and Western blotting methods were applied in detected the expression of miR-155 and AGTR1.Results The expression of miR-155 in placental tissues showed an increasing trend with the PE deteriorated,the expression quantity of control group,PE group and severe PE group were(1.18±0.12),(1.57±0.07)and(1.98±0.10)respectively and there was statistical difference on the whole(F=384.2,P<0.001).The expression of AGTR1 were displayed a decreasing trend with the situation deteriorated,the expression of these groups were(0.86±0.08),(0.57±0.04)and(0.29±0.07),and there was statistical difference on the whole(F=99.4,P<0.001).Conclusion The expressed situation of miR-155 and AGTR1 were varied in placental tissues of patients with preeclampsia according to extent.In the placenta of patients with preeclampsia,miR-155 is expressed at higher levels,while AGTR1 is expressed at lower levels.With the worse of disease,the expression of miR-155 increased while AGTR1 decreased.So we speculated that miR-155 may influence pathogenesis of preeclampsia by regulating AGTR1.
