摘要
目的探讨肌电相同的喉内外肌NGF及Trk A表达的区别,以期使用喉外肌替代病废的喉内肌提供理论依据。方法 10只健康成年犬,取环杓后肌,及与其肌电类同的胸骨甲状肌,颏舌骨肌,取环杓侧肌,及与其肌电类同的下颌舌骨肌,甲状舌骨肌。用蛋白免疫印迹法(Western blot)在蛋白水平检测神经生长因子(NGF)及Trk A的表达;用荧光定量PCR在基因水平检测NGF及Trk A的表达。结果 WB及荧光定量PCR检测显示,NGF方面胸骨甲状肌高于环杓后肌(P<0.05),颏舌骨肌与环杓后肌比较无显著性差异(P>0.05),甲状舌骨肌高于环杓侧肌(P<0.05),下颌舌骨肌与环杓侧肌比较无显著性差异(P>0.05);Trk A水平上胸骨甲状肌高于环杓后肌(P<0.05),颏舌骨肌与环杓后肌比较无显著性差异(P>0.05),下颌舌骨肌高于环杓侧肌(P<0.05),甲状舌骨肌与环杓侧肌比较无显著性差异(P>0.05)。结论 NGF表达方面,颏舌骨肌与环杓后肌水平相同,下颌舌骨肌与环杓侧肌水平相同;Trk A表达方面,颏舌骨肌与环杓后肌水平相同,甲状舌骨肌与环杓侧肌相同。
Objective To explore the difference in the expression of NGF and Trk A in the internal and external laryngeal muscles with the same EMG,providing a theoretical basis for replacing the defective internal laryngeal muscle with the external laryngeal muscle.Methods The mylohyoid muscles,geniohyoid muscles,sternothyroid muscles,thyrohyoid muscles,lateral cricoaryteoid muscle,and posterior cricoaryteoid muscle were taken out from ten healthy adult dogs.Western blot was used to detect the expression of nerve growth factor(NGF)and Trk A at the protein level;the expression of NGF and Trk A was detected at the gene level by fluorescent quantitative PCR.Results WB and fluorescent quantitative PCR showed that,In terms of NGF,the sternohyoid muscle is higher than the posterior cricoaryteoid muscle(P<0.05),there is no significant difference between the geniohyoid muscle and the posterior cricoaryteoid muscle(P>0.05).The thyrohyoid muscle is higher than the lateral cricoaryteoid muscle(P<0.05),and there is no significant difference between the mylohyoid muscles and lateral cricoaryteoid muscle(P>0.05);the sternohyoid muscle is higher than the posterior cricoaryteoid muscle at the Trk A(P<0.05),there was no significant difference between the geniohyoid muscle and the posterior cricoaryteoid muscle (P>0.05),the mylohyoid muscles is higher than the lateral cricoary-teoid muscle(P<0.05),There was no significant difference between the thyrohyoid muscle and the lateral cricoaryteoid muscle(P>0.05).Conclusion In terms of the expression of NGF,the geniohyoid muscle and the posterior cycloid muscleare at the same level,and the The mylohyoid muscles is at the same level as the lateral cricoaryteoid muscle.For the expression of Trk A,the geniohyoid muscle and the posterior cricoaryteoid muscle are at the same level,and the thyrohyoid muscles is the same as lateral cricoaryteoid muscle.
