纹党和红芪加工废弃物中活性多糖对衰老小鼠学习记忆能力和抗疲劳作用的影响 被引量:4

摘要 【目的】为探讨纹党和红芪加工废弃物中活性多糖对衰老小鼠学习记忆能力和抗疲劳作用的影响.【方法】以小鼠体质量和血清中超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)活性及丙二醛(malondialdehyde,MDA)含量为检测指标,筛选出纹党多糖(Codonopisis pilosula polysaccharides,CPP)和红芪多糖(Hedysarum polybotrys polysaccharides,CHP)的最佳剂量是400 mg/kg;再以0.5 mL/10 g剂量颈部皮下注射5%的D-半乳糖(D-galactose)溶液6周后,构建小鼠亚急性衰老模型,检测小鼠体质量及血清中SOD和CAT的活性和丙二醛MDA含量的变化,确定是否建模成功.将建模成功的小鼠随机分为CK(0)、衰老模型组、纹党多糖、红芪多糖剂量处理组和阳性处理组,检测2种多糖对衰老小鼠走迷宫所用的学习记忆时间、游泳时间、爬杆时间和耐缺氧时间的变化.【结果】衰老小鼠分别用400 mg/kg剂量的CCP和CHP连续灌胃给药4周后,衰老小鼠体质量恢复明显,小鼠走迷宫所用的学习记忆时间逐渐缩短,小鼠游泳时间、爬杆时间和耐缺氧时间逐渐延长,且CCP效果优于CHP,两者联用效果最好,与VE组无显著差异.【结论】纹党和红芪加工废弃物中活性多糖能恢复衰老小鼠的学习记忆能力,对衰老小鼠的抗疲劳作用和常压耐缺氧有显著的延长作用. 【Objective】To investigate the effects of active polysaccharides from Codonopisis pilosula and Hedysarum polybotrys processing wastes on learning and memory ability and antifatigue in aging mice.【Method】Firstly,the activities of superoxide dismutase(SOD)and catalase(CAT)and the content of malondialdehyde(MDA)in serum and body weight of mice were examined,which were taken as the detection indexes,and the optimal dosage of C.pilosula polysaccharides(CPP)and H.polybotrys polysaccharides(CHP)were selected as 400 mg/kg·bw.Secondly,subacute aging model of mice was established by subcutaneous injected in the neck 5%D-galactose solution at the dose of 0.5 mL/10g for 6 weeks were measured to determine whether the model was successful.The mice were randomly divided into CK(0),aging model group,CCP and CHP dose treatment group and positive treatment group(VE 170 mg/kg),the changes of learning and memory time,swimming time,pole climbing time and oxygen tolerance time of aging mice were detected.【Result】The weight of aging mice recovered significantly after continuous intragastric administration with 400 mg/kg CCP and CHP for 4 weeks,the learning and memory time of mice in maze were gradually shortened,swimming time,pole climbing time and hypoxia tolerance time of mice were gradually prolonged,and the effect of CCP was better than CHP,at the same time,there was no difference compared with VE treatment group.【Conclusion】The CPP and CHP could restore the learning and memory ability of aging mice,and significantly prolong the antifatigue effect and antihypoxia effect under normal pressure.
