肺炎颗粒成型工艺的优化及其对小鼠的镇咳作用 被引量:4

摘要 目的优化肺炎颗粒成型工艺,并评价其镇咳作用。方法在单因素试验基础上,以浸膏粉与稀释剂(乳糖)比例、润湿剂(乙醇)体积分数、润湿剂用量为影响因素,成型率为评价指标,正交试验优化成型工艺。采用小鼠氨水引咳实验,考察肺炎颗粒的镇咳作用。结果最佳条件为浸膏粉与稀释剂比例1∶1,润湿剂体积分数90%,润湿剂用量15%~20%,矫味剂(三氯蔗糖)用量0.08%,成型率96.4%,吸湿率1.91%,休止角7.12°,含水量4.62%。与模型组比较,高(9.4 g/kg)、中(4.7 g/kg)、低(2.4 g/kg)剂量肺炎颗粒组咳嗽潜伏期延长(P<0.05,P<0.01),咳嗽次数减少(P<0.01)。结论该方法合理可行,所得肺炎颗粒性质稳定,吸湿性低,溶化性好,并具有明显镇咳作用。 AIM To optimize the forming process for granules for pneumonia and to evaluate their antitussive effect.METHODS With extract powder-diluent(lactose)ratio,wetting agent(ethanol)concentration and wetting agent consumption as influencing factors,forming rate as an evaluation index,the forming process was optimized by orthogonal test on the basis of single factor test.The antitussive effect was investigated by ammonia-induced mouse cough test.RESULTS The optimal conditions were determined to be 1∶1 for extract powder-diluent ratio,90%for wetting agent concentration,15%-20%for wetting agent consumption,and 0.08%for corrigent(sucralose)consumption,the forming rate,moisture adsorption rate,angle of repose and moisture content were 96.4%,1.91%,7.12°and 4.62%,respectively.Compared with the model group,high-dose(9.4 g/kg),medium-dose(4.7 g/kg)and low-dose(2.4 g/kg)granules for pneumonia groups demonstrated prolonged cough incubation period(P<0.05,P<0.01)and reduced cough frequency(P<0.01).CONCLUSION The granules for pneumonia obtained by this reasonable and feasible method exhibit stable properties,low moisture absorption,good dissolubility and obvious antitussive effect.
