参芎葡萄糖注射液对CoCl_(2)诱导的H9c2细胞凋亡的影响 被引量:3

摘要 目的探讨参芎葡萄糖注射液对氯化钴(CoCl_(2))诱导的H9c2心肌细胞凋亡的拮抗作用及其机制。方法体外培养H9c2细胞,利用CoCl_(2)建立细胞凋亡模型;MTS法检测细胞存活率;流式细胞术检测细胞凋亡率;蛋白免疫印迹法(Western blot)检测c-Jun、p-c-Jun、Bcl-2、Bax和Cleaved caspase 3的表达情况。结果CoCl_(2)浓度在600μmol/L处理24 h的情况下可建立H9c2细胞凋亡模型;不同浓度的SGI预处理后,可明显增加细胞存活率,降低细胞凋亡率,下调蛋白c-Jun磷酸化水平、Bax和Cleaved caspase 3表达水平,上调蛋白Bcl-2表达水平。结论SGI预处理对CoCl_(2)诱导H9c2细胞凋亡具有拮抗作用,其机制可能与其抑制c-Jun通路有关。 AIM To explore the antagonistic effect of Shenxiong Glucose Injection(SGI)on CoCl_(2)-induced H9c2 cells apoptosis and its mechanism.METHODS H9c2 cells cultured in vitro were induced into ap-optotic models using CoCl_(2).H9c2 cells were subjected to the detection of cell survival by MTS;the determination of cell apoptosis by flow cytometry;the identification of protein expressions of c-Jun,p-c-Jun,Bcl-2,Bax and Cleaved caspase 3 by Western blot.RESULTS H9c2 cells apoptosis models were established under 24 h 600μmol/L CoCl_(2) treatment.SGI retreatment at different concentrations significantly increased the cell survival rate,reduced the ap-optosis rate,and significantly reduced the protein expression levels of p-c-Jun/c-Jun,Bax and Cleaved caspase 3,and up-regulated protein Bcl-2 expression level.CONCLUSION SGI pretreatment protects H9c2 cells from CoCl_(2)-induced apoptosis,and its mechanism may be associated with the c-Jun pathway inhibition.
