ApoE基因多态性与缺血性脑卒中合并2型糖尿病的相关性分析

Correlation between apolipoprotein E genetic polymorphism and ischemic cerebral stroke complicated by type 2 diabetes mellitus

目的分析ApoE基因多态性与缺血性脑卒中合并2型糖尿病的相关性。方法采用病例-对照的方法,选取2018年6月至2019年2月收治的512例缺血性脑卒中患者,其中164例合并2型糖尿病患者(T2DM组),348例不合并2型糖尿病患者(NC组)。用PCR扩增和特异性核酸探针基因芯片检测ApoE基因型。数据用SPSS21.0进行检验并使用logistic回归分析。结果ApoE基因E2/E2基因型3例(0.59%)、E2/E3基因型78例(15.23%)、E2/E4基因型9例(1.76%)、E3/E3基因型331例(64.65%)、E3/E4基因型88例(17.19%)、E4/E4基因型3例(0.59%);等位基因ApoE3分布最多占比80.86%,ApoE4其次占比10.06%,ApoE2分布最少占比9.08%。ApoE基因型频率符合Hardy-Weinberg遗传平衡定律(P>0.05)。T2DM组TG水平比NC组高,而AST、CK和HDL-C水平显著低于NC组(P<0.05)。ApoE基因型在T2DM组和NC组比较差异无统计学意义(P>0.05)。经logistic回归分析发现ApoE基因多态性不是缺血性脑卒中患者合并2型糖尿病的危险因素(P>0.05)。结论ApoE基因不是缺血性脑卒中患者合并2型糖尿病的危险因素。

Objective To investigate the correlation between apolipoprotein E genetic polymorphism and ischemic cerebral stroke complicated by type 2 diabetes mellitus.Methods A total of 512 patients with ischemic cerebral stroke who were admitted and treated in our hospital from June 2018 to February 2019 were enrolled in the study,of whom,there were 164 patients with type 2diabetes mellitus(T2DM group)and 348 patients without type 2 diabetes mellitus(NC group).The ApoE genotype was detected by PCR amplification and specific nucleic acid probe gene chip.The data were tested by means of SPSS21.0 software and Logistic regression analysis.Results There were 3 cases of ApoE E2/E2 genotype(0.59%),78 cases of E2/E3 genotype(15.23%),9 cases of E2/E4 genotype(1.76%),331 cases of E3/E3 genotype(64.65%),88 cases of E3/E4 genotype(17.19%),and 3 cases of E4/E4 genotype(0.59%).The distribution of allele ApoE3 accounted for 80.86%at most,ApoE410.06%,and ApoE29.08%at least.ApoE genotype was consistent with Hardy-Weinberg’s law of genetic equilibrium(P>0.05).Moreover the TG levels in T2DM group were significantly higher than those in NC group,however,the levels of AST,CK and HDL-C were significantly lower than those in NC group(P<0.05).There was no significant difference in ApoE genotype between T2DM group and NC group(P>0.05).Logistic regression analysis showed that ApoE gene polymorphism was not a risk factor for the patients with ischemic cerebral stroke complicated by type 2 diabetes mellitus(P>0.05).Conclusion ApoE gene is not the risk factor for the patients with ischemic cerebral stroke complicated by type 2 diabetes mellitus.
