益肾泄浊法对糖尿病肾病大鼠TGF-β1、CTGF蛋白及基因表达的影响 被引量:3

摘要 目的:以转化生长因子-β1(TGF-β1)、结缔组织生长因子(CTGF)蛋白为基础,探讨益肾泄浊法对糖尿病肾病大鼠的影响。方法:采用链脲佐菌素(STZ)建立大鼠糖尿病(Diabetes mellitus, DM)模型并给予不同剂量益肾泄浊法干预,进行生化检查、蛋白免疫印迹试验(Western blot)和实时荧光定量聚合酶链式反应(Real-time Polymerase Chain Reaction)。结果:与模型组相比,益肾泄浊法组可以降低DM组大鼠肾指数(KI)和尿肌酐(Ucr),益肾泄浊法高、中、低剂量组均能使TGF-β1、CTGF蛋白和mRNA的表达量减少。结论:益肾泄浊法可以减少TGF-β1和CTGF的表达,提示其可通过激活TGF-β1/CTGF信号通路从而达到治疗糖尿病肾病的作用。 Objective: To explore the effect of Kidney-tonifying and Turbidity-discharging Method for diabetic nephropathy rats on the basis of transforming growth factor-β1(TGF-β1) and connective tissue growth factor(CTGF) protein.Methods: The rat DM model was established by streptozotocin(STZ) and treated with different doses of Kidney-tonifying and Turbidity-discharging Method.Performed biochemical examination, Western blot detection and Real-time PCR detection.Results: Compared with the model group, the kidney-tonifying and turbid-relieving method group can reduce the renal index(KI) and urinary creatinine(Ucr) of rats in the DM group.The high, middle and low dose groups of Kidney-tonifying and Turbidity-discharging Method could reduce the expression of TGF-β1,CTGF protein and mRNA.Conclusion: Kidney-tonifying and Turbidity-discharging Method can reduce the expression of TGF-β1 and CTGF,suggesting that Kidney-tonifying and Turbidity-discharging Method can treat diabetic nephropathy by activating TGF-β1/CTGF signal pathway.
