Screening and expression analysis of latent pulmonary tuberculosis biomarkers in high risk medical institutions

摘要 目的筛选健康人群(HC)和潜伏性感染肺结核患者(LTBI)的血浆中差异表达的microRNAs(miRNAs),探讨microRNAs诊断潜伏性肺结核的应用价值。方法利用QFT实验筛选HC组与LTBI组各22例。提取外周抗凝血分离血浆中总RNA,利用q PCR实验检测五个miRNA的相对表达量。结果在LTBI组中miR-144-5p、miR-144-3p显著高于HC组,分别为HC组的2.31倍和1.67倍,差异有统计学意义(P<0.05)。ROC曲线分析说明miR-144-5p、miR-144-3p的特异性分别为81.80%、72.70%,敏感性分别为65.90%、75.00%。结论miR-144-5p、miR-144-3p具有作为潜伏性肺结核筛查的生物标记物的应用前景。 Objective To screen microRNAs(miRNAs)differentially expressed in plasma of healthy people(HC)and patients with latent pulmonary tuberculosis(LTBI),and to explore the application value of microRNAs in the diagnosis of latent pulmonary tuberculosis.Methods 22 cases of HC group and LTBI group were screened by QFT test.Total RNA was extracted from plasma from peripheral anticoagulant blood,and the relative expression levels of five miRNAs were detected by q PCR assay.Results miR-144-5 p and miR-144-3 p in LTBI group were significantly higher than those in HC group,which were 2.31 times and 1.67 times of those in HC group,respectively,with statistical significance(P<0.05).ROC curve analysis showed that the specificities of miR-144-5 p and miR-144-3 p were 81.80%and 72.70%,respectively,and the sensitivities were 65.90%and 75.00%.Conclusion miR-144-5 p and miR-144-3 p have potential application as biomarkers for screening latent tuberculosis.
