基于网络药理学方法探讨益气活血方治疗糖尿病合并动脉粥样硬化的作用机制 被引量:6

摘要 目的运用网络药理学方法探讨益气活血方治疗糖尿病合并动脉粥样硬化的潜在作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP),筛选出益气活血方的活性成分并进行靶点预测,利用Genecards、OMIM、Drugbank数据库获得糖尿病合并动脉粥样硬化疾病靶点,应用Cytoscape 3.7.1软件,构建益气活血方成分靶点-疾病靶点网络;利用Metascape进行基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析预测益气活血方治疗糖尿病合并动脉粥样硬化的作用机制。结果筛选出益气活血方有效活性成分28个、药物靶点122个、糖尿病合并动脉粥样硬化靶点1275个,最终获得关键靶点191个、KEGG通路264条。结论初步揭示了益气活血方干预糖尿病合并动脉粥样硬化的活性成分和潜在靶点,为其在临床的进一步应用和深入研究提供了依据和思路。 Objective To explore the potential mechanism of Yiqi Huoxue Decoction in the treatment of diabetes complicated with atherosclerosis by network pharmacology.Methods The active ingredients of Yiqi Huoxue Decoction and target predictions were screened out through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The diabetes and atherosclerotic disease targets were obtained by Genecards,OMIM,and Drugbank databases.The component target-disease network of Yiqi Huoxue Decoction were constructed by Cytoscape 3.7.1 software.The mechanism of Yiqi Huoxue Decoction in treating diabetes complicated with atherosclerosis were predicted by GO and KEGG enrichment analysis.Results Twenty-eight active ingredients of Yiqi Huoxue Decoction,122 drug targets,1275 diabetes and atherosclerosis targets were screened out,and finally 191 key targets and 264 KEGG pathways were obtained.Conclusion The active ingredients and potential targets of Yiqi Huoxue Decoction for intervention of diabetes mellitus complicated with atherosclerosis are predicted,which provides basis and ideas for its further clinical application and further research.
