摘要
天然产物(-)-Sparteine作为喹诺里西丁类生物碱,具有显著的生理活性,一直是新药研发的热点。并且,(-)-Sparteine作为优良的手性配体,在手性化学领域也扮演着重要角色。但是,从植物中提取(-)-Sparteine步骤繁琐,效率较低,不适用于工业化生产,目前市面上,(-)-Sparteine也难以购得。因此,研究(-)-Sparteine的合成是极为必要的。文章综述了关于天然产物(-)-Sparteine合成的研究进展,从生物合成路径、半合成和全合成方法的角度进行了分类阐述,有利于今后对(-)-Sparteine进行工业化生产,改善其来源稀缺的现状。
The natural product(-)-Sparteine,as one of the quinolizidine alkaloids,possesses significant physiological effects and has always been attractive in the research and development of new drugs.Additionally,(-)-Sparteine was also broadly applied as a chiral ligand in diverse catalytic asymmetric reactions,greatly promoting the advancement of chiral chemistry.However,extracting(-)-Sparteine from the natural plants can not only be complicated,but also inefficient,not practical for industrial production.At present,(-)-Sparteine is even unavailable in market.Thus,it's urgent to explore the synthesis of(-)-Sparteine in order to make up for its shortage.Herein,the advances on(-)-Sparteine synthesis were reviewed from the aspect of biosynthesis,semi-synthesis and total synthesis.It is remarkable for developing the industrial production of(-)-Sparteine in order to improve the current situation of(-)-Sparteine scarcity.
