猪源马链球菌兽疫亚种强毒株感染小鼠模型建立

Establishment of A Mouse Model Infected with A Virulent Strain of Streptococcus Equi Subsp Zooepidemicus from Swine

摘要 目的:通过马链球菌兽疫亚种强毒株感染小鼠致病,使其链球菌感染并且采集得到阳性血清。方法:共有20只8周龄ICR清洁小鼠,其中1组为对照组,每1只注射剂量为0.7 mL PBS,另1组感染组注射剂量为0.7 mL强毒株菌液,每隔2 h观察1次。采集感染症状明显且濒临死亡的小鼠血液,随后进行剖检,分离肝脏组织细菌。结果:攻毒小鼠不喜动,毛发粗乱,眼部炎症明显,几近失明,呼吸困难,攻毒组小鼠有2只濒临死亡,其余小鼠均有明显临床症状,对照组小鼠未见任何异常,从肝脏成功分离得到链状分布的球状菌,可见小鼠攻毒成功,由此可见成功得到马链球菌兽疫亚种感染小鼠阳性血清,可用于研究细菌感染产生抗体的检测分析,以及为研究小鼠免疫机制等方面提供原材料,为后续科研工作奠定基础。结论:成功建立猪源马链球菌兽疫亚种强毒株感染小鼠模型。 Purpose:to infect mice with a virulent strain of streptococcus equi subsp.zooepidemicus to make them infected with Streptococcus and collect positive sera.Methods:a total of 208-week-old ICR clean mice,of which one group was the control group,each injected with a dose of 0.7 mL PBS,and the other group in the infection group was injected with a dose of 0.7 mL of virulent strain,every 2 h observe once.Blood was collected from mice with obvious symptoms of infection and dying,and then autopsy was performed to isolate bacteria from liver tissue.Results:the challenged mice were unhappy,had coarse hair,obvious eye inflammation,almost blindness,and difficulty breathing.Two mice in the challenge group were dying,and the remaining mice had obvious clinical symptoms.The mice in the control group no abnormality was seen.The chain-like cocci were successfully isolated from the liver.It can be seen that the mice were successfully challenged.It can be seen that the positive serum of mice infected with streptococcus equi subsp.zooepidemics was successfully obtained,which can be used to study the detection of antibodies produced by bacterial infections.Analyze and provide raw materials for studying the immune mechanism of mice,laying the foundation for subsequent scientific research.Conclusion:successfully established a mouse model of swine-derived streptococcus equi subsp zooepidemic strain infection.
