摘要
生物芯片是生物学领域的最新技术,其中抗体常被固定在蛋白质芯片上作为研究蛋白质组学的工具,但抗体不稳定、难以针对毒性靶标等缺点限制了抗体芯片的发展,适配体芯片则提供了替代技术来规避抗体芯片的限制。本文回顾了生物芯片的发展以区分DNA芯片和适配体芯片,并从制备方法、检测手段及优化策略三个方面展开对适配体芯片的介绍。最后对适配体芯片技术的最新进展进行综述,并阐述了适配体芯片的局限性和未来前景。
Biochips are the latest technology in the field of biology,in which antibodies are often immobilized on protein microarrays as tools for proteomics study.However,the instability of antibodies and difficulty in targeting toxic targets limit the development of antibody microarrays,while the aptamer microarrays provide a new method to circumvent these limitations as alternative.This paper reviewed the development of biochips to distinguish DNA chips and aptamer microarrays,and introduced the aptamer microarray from three aspects,including the preparation method,detection method and optimization strategy of the aptamer microarray.Finally the latest developments of aptamer microarrays are reviewed to shed light on current limitations and future prospects of aptamer microarrays.
