摘要
目的分析人血红素加氧酶-1(hHO-1)基因启动子区序列特点,转录因子结合位点种类及CpG岛位置。方法利用美国国立生物技术信息中心(NCBI)数据库获取hHO-1基因序列和启动子序列,SoftBerry分析hHO-1基因核心启动子位置,PROMO和Gene Regulation分析转录因子结合位点种类,MethPrimer 2.0和CpGFinder预测CpG岛位置。结果hHO-1基因位于22q12.3,全长13112 bp。转录起始点上游约1995 bp范围内为启动子区域,hHO-1启动子区含79种转录因子结合位点和1个CpG岛。结论hHO-1基因启动子能预测出多种转录因子结合位点,可为更深入地研究hHO-1基因的调控机制及其在抗逆方面的潜在作用提供理论基础。
Objective To analyze the sequence characteristics,kinds of transcription factors binding sites and location of CpG island of human heme oxygenase 1(hHO-1)gene promoter region.Methods The sequences of hHO-1 gene and its promoter were obtained from the NCBI database.The position of hHO-1 gene core promoter was analyzed by SoftBerry.The type of transcription factor binding sites was analyzed by PROMO and Gene regulation.The position of CpG island was predicted by MethPrimer 2.0 and CpGFinder.Results The hHO-1 gene was located at 22q12.3,and the full length was 13112 bp.The promoter region was about located in the 1995 bp upstream of transcription initiation site.There might be 79 species of transcription factor binding sites and 1 CpG islands in the promoter region of the hHO-1 gene.Conclusion The hHO-1 gene promoter can predict many kinds of transcription factor binding sites,which provides a theoretical basis for further research on the regulation mechanism of hHO-1 gene and its potential role of stress resistance.
