摘要
目的探讨辣椒素通过调节GRP78触发内质网应激对人急性髓系白血病HL-60细胞凋亡的影响及机制。方法CCK-8筛选辣椒素的最佳作用浓度;Wright-Giemsa染色观察HL-60细胞的形态;Western blot检测GRP78蛋白的表达量;Annexin V-FITC/PI双染法检测HL-60细胞的凋亡率。结果辣椒素的最佳作用浓度为40、60和80μmol/L;辣椒素可使HL-60细胞出现染色质浓集,呈半月状或指环状等变化,且随着辣椒素浓度的增加,HL-60细胞的数目逐渐减少;经不同浓度辣椒素处理后的HL-60细胞,其细胞凋亡率逐渐增加(P<0.001);GRP78蛋白的表达量随着辣椒素浓度的增加而增加(P<0.01),加入Salubrinal预处理后,GRP78蛋白的表达明显减少(P<0.01),HL-60细胞的凋亡率也下降(P<0.01);辣椒素增加的GRP78蛋白的表达量在转染shGRP78后下调(P<0.001),HL-60细胞的凋亡率也下降(P<0.01)。结论辣椒素显著降低HL-60细胞的细胞活力,促进HL-60细胞的凋亡,其可能与调节GRP78触发内质网应激有关。
Objective To investigate the effect and mechanism of capsaicin on the apoptosis of human acute myeloid leukemia HL-60 cells by triggering endoplasmic reticulum stress through regulating GRP78.Methods CCK-8 was used to screen the best concentration of capsaicin;Wright-Giemsa staining was used to observe the morphology of HL-60 cells;Western blot was used to detect the expression of GRP78 protein;Annexin V-FITC/PI double staining method was used to detect the apoptosis rate of HL-60 cells.Results The optimal concentration of capsaicin was:20,40 and 60μmol/L;capsaicin could cause HL-60 cells to exhibit chromatin concentration,showing a half-moon-shaped or finger ring-like change,and as the concentration of capsaicin increases,the number of HL-60 cells gradually decreased;after treatment with different concentrations of capsaicin,the apoptosis rate of HL-60 cells gradually increased(P<0.001);the expression of GRP78 protein increased with the increase of capsaicin concentration(P<0.01),and after adding Salubrinal pretreatment,the expression of GRP78 protein was significantly reduced(P<0.01),and the apoptosis rate of HL-60 cells also decreased(P<0.01);the increased expression of GRP78 protein by capsaicin was down-regulated after transfection with shGRP78(P<0.001),and the apoptosis rate of HL-60 cells also decreased(P<0.01).Conclusion Capsaicin significantly reduces the viability of HL-60 cells and promotes the apoptosis of HL-60 cells,which may be related to the regulation of GRP78 to trigger endoplasmic reticulum stress.
