洋甘菊提取物对多囊卵巢综合征大鼠子宫内膜组织、胰岛素抵抗、瘦素和血脂水平的影响 被引量:7

Effects of chamomile extract on endometrial tissue,insulin resistance,leptin and blood lipid levels in rats with polycystic ovary syndrome

摘要 目的探讨洋甘菊提取物对多囊卵巢综合征大鼠子宫内膜组织、胰岛素抵抗、瘦素和血脂水平的影响。方法Wistar大鼠分为假手术组(sham组)、多囊卵巢综合征组(PCOS组)、洋甘菊提取物低剂量组(PCOS+CHA-L组)和洋甘菊提取物高剂量组(PCOS+CHA-H组);测量各组大鼠体重和子宫重量,并计算子宫指数;放射免疫分析法检测各组大鼠血清黄体生成素、卵泡刺激素、睾丸激素、孕酮和雌二醇的表达水平;HE染色观察各组大鼠子宫内膜组织和胰腺组织的病理损伤情况;葡萄糖氧化酶法检测各组大鼠血清中葡萄糖的表达水平;放射免疫分析法检测各组大鼠胰岛素的表达水平,计算胰岛素抵抗指数;ELISA法检测各组大鼠血清中瘦素、三酰甘油、胆固醇、高密度脂蛋白和低密度脂蛋白的表达水平。结果洋甘菊提取物使大鼠的体重和子宫指数均下降(P<0.05),黄体生成素表达水平和睾丸激素表达水平均下降(P<0.05),卵泡刺激素表达水平和雌二醇表达水平均上升(P<0.05);洋甘菊提取物使子宫内膜腺体正常,乳头状排列,炎性细胞减少,也使胰岛组织形态和结构趋于正常,细胞排列紧密;与PCOS组相比,PCOS+CHA-L组和PCOS+CHA-H组大鼠血清胰岛素含量、葡萄糖含量和胰岛素抵抗指数降低(P<0.05);洋甘菊提取物使大鼠血清瘦素水平下降(P<0.05),三酰甘油、胆固醇和低密度脂蛋白的含量明显下降(P<0.05),高密度脂蛋白含量增加(P<0.05)。结论洋甘菊提取物可通过降低体重、子宫指数和胰岛素抵抗指数,调节性激素、瘦素和血脂水平,从而改善PCOS大鼠的生殖功能。 Objective To investigate the effect of chamomile extract on endometrial tissue,insulin resistance,leptin and blood lipid levels in rats with polycystic ovary syndrome.Methods Wistar rats are divided into sham operation group(sham group),polycystic ovary syndrome group(PCOS group),low-dose group of chamomile extract(PCOS+CHA-L group)and high-dose group of chamomile extract(PCOS+CHA-H Group).The body mass and uterine mass of rats in each group were recorded,and the uterine index was calculated.Radioimmunoassay to detect the expression levels of serum luteinizing hormone,follicle stimulating hormone,testosterone,progesterone and estradiol in each group of rats.HE staining was used to observe the pathological damage of the endometrial tissue and pancreatic tissues of rats in each group.Glucose oxidase method was used to detect the expression level of glucose in the serum of each group of rats.Radioimmunoassay was used to detect the expression level of insulin in each group,and the insulin resistance index was calculated according to the formula.The expression levels of leptin,triglyceride,cholesterol,high-density lipoprotein and low-density lipoprotein in serum of rats in each group were detected by ELISA.Results Chamomile extract decreased the body mass and uterine index of rats(P<0.05),decreased the expression levels of luteinizing hormone and testosterone(P<0.05),and increased the expression levels of follicle stimulating hormone and estradiol(P<0.05).The chamomile extract made the endometrial glands normal,papillary,reduced inflammatory cells,and also made the islet tissue morphology and structure turn to normal,and the cells closely arranged.Compared with the PCOS group,the serum insulin content,glucose content and insulin resistance index of the PCOS+CHA-L group and PCOS+CHA-H group decreased(P<0.05).Chamomile extract decreased the serum leptin level of rats(P<0.05),decreased the content of triglyceride,cholesterol and low-density lipoprotein(P<0.05),and increasd the content of high-density lipoprotein(P<0.05).Conclusion Chamomile extract can improve the reproductive function of PCOS rats by reducing body weight,uterine index and insulin resistance index and regulating sex hormones,leptin and blood lipid levels.
