人参皂苷Rb1对系统性红斑狼疮患者外周血T淋巴细胞体外活化的影响 被引量:3

Effect of ginsenoside Rb1 on activation of serum T lymphocytes in vitro in patients with systemic lupus erythematosus

摘要 目的探讨人参皂苷Rb1对系统性红斑狼疮(SLE)患者外周血T淋巴细胞的免疫调节作用。方法收集处于非活动期SLE患者(30例)和健康对照人群(30例)的外周血,采用免疫磁珠法分离CD3^(+)T淋巴细胞,分为空白组、刀豆球蛋白A(ConA)组、ConA^(+)Rb1组。3H-TdR渗入法检测各组细胞增殖,并采用流式细胞术检测CD3^(+)CD69^(+)T细胞、CD4^(+)CD25^(+)T细胞和CD4^(+)CD25^(+)FoxP3^(+)T细胞比例,ELISA法检测T淋巴细胞培养上清中白细胞介素4(IL-4)和干扰素-γ(IFN-γ)水平。结果当人参皂苷Rb1浓度为10μmol/L时,可抑制外周血T淋巴细胞的增殖(抑制率为40.87%±9.82%、53.94%±9.12%),且对SLE人群外周血T淋巴细胞增殖的抑制作用高于健康对照人群(P<0.05);且可抑制CD3^(+)CD69^(+)T细胞比例(31.28%±5.32%vs.33.64%±5.79%,P>0.05)。此外,健康对照组和SLE组外周血T淋巴细胞CD4^(+)CD25^(+)T细胞百分比(24.38%±5.45%、16.29%±3.18%)和CD4^(+)CD25^(+)FoxP3^(+)T细胞百分比(32.76%±7.43%、26.35%±5.14%)较ConA组显著增加(P<0.05)。人参皂苷Rb1还可促使T淋巴细胞分泌IFN-γ,同时抑制IL-4分泌。结论人参皂苷Rb1可抑制ConA刺激的SLE患者外周血T淋巴细胞增殖活性,同时增加CD4^(+)CD25^(+)T细胞比例,维持Th1/Th2免疫平衡,有望成为SLE的治疗药物。 Objective To investigate the regulating effect of ginsenoside Rb1 on serum T lymphocytes in patients with systemic lupus erythematosus(SLE).Methods The plasma samples from 30 patients with inactivated SLE and 30 healthy volunteers in our hospital were collected for the study,which were divided into blank group,ConA group and ConA^(+)Rb1 group.CD3^(+)T cells were isolated by immunomagnetic beads.3H-TdR infiltration method was used to detect the proliferation of T lymphocytes.The ratio of CD3^(+)CD69^(+)T cells,CD4^(+)CD25^(+)T cells and CD4^(+)CD25^(+)FoxP3^(+)T cells was detected by flow cytometry.The levels of interleukin4(IL-4)and interferon-γ(IFN-γ)in the supernatant of T lymphocytes were detected by ELISA.Results When the concentration of ginsenoside Rb1 was 10μmol/L,the proliferation of peripheral blood T lymphocytes was inhibited(inhibition rate:40.87%±9.82%and 53.94%±9.12%),and its inhibitory effect on peripheral blood T lymphocyte proliferation in SLE population was higher than that in healthy control group(P<0.05),and the proportion of CD3^(+)CD69^(+)T cells(31.28%±5.32%vs.33.64%±5.79%,P>0.05)was also inhibited.In addition,the percentage of CD4^(+)CD25^(+)T cells(24.38%±5.45%and 16.29%±3.18%)and the percentage of CD4^(+)CD25^(+)Foxp3^(+)T cells(32.76%±7.43%and 26.35%±5.14%)of peripheral blood T lymphocytes in healthy control group and SLE group were significantly higher than those in ConA group(P<0.05).Ginsenoside Rb1 could also promote the secretion of IFN-γand inhibit the secretion of IL-4.Conclusion Ginsenoside Rb1 can inhibit the proliferation of T lymphocytes in peripheral blood of SLE patients stimulated by ConA,increase the proportion of CD4^(+)CD25^(+)T cells and maintain Th1/Th2 immune balance,which is expected to be a therapeutic drug for SLE.
