鼻咽癌高癌家系癌患者外周血淋巴细胞S100A9表达分析

Analysis of S100A9 expression of peripheral blood lymphocytes in patients with nasopharyngeal carcinoma in high familial

目的探索鼻咽癌高癌家系癌患者外周血淋巴细胞S100A9差异性表达情况及其临床意义。方法收集高癌家系鼻咽癌患者、散发鼻咽癌患者及正常对照组外周血各22例,分别抽取患者及对照组病人外周静脉血10ml,提取外周血淋巴细胞进行S100A9蛋白分析,采用Western blot、RT-PCR分别检测三组研究对象S100A9蛋白、S100A9mRNA表达情况。结果鼻咽癌高癌家系癌患者及鼻咽癌散发癌患者及正常对照组S100A9 mRNA表达分别为-3.886±1.663、-3.421±1.110、-4.031±1.586,三组比较P>0.05,差异无统计学意义。S100A9蛋白表达分别为0.518±0.353、0.481±0.344、0.245±0.202,三组比较P<0.05,差异具有统计学意义。结论鼻咽癌高癌家系癌患者及散发鼻咽癌患者外周血淋巴细胞S100A9蛋白表达与正常人比存在差异性,有望成为鼻咽癌早期筛查指标。

Objective To explore the differential expression of S100A9 in peripheral blood lymphocytes in patients with nasopharyngeal carcinoma in high-cancerous family and its clinical significance.Methods It had been collected of peripheral blood lymphocytes in three group peoples including familial nasopharyngeal carcinoma,distribution nasopharyngeal carcinoma and normal people,each group had 22 cases.Peripheral venous blood 10ml samples were extracted from patients and control group respectively,in which peripheral lymphocytes were extracted for proteomic analysis.western blot,RT-PCR were used to detect the expression of S100A9 protein and S100A9 mRNA in three groups of subjects.Results High familial nasopharyngeal carcinoma patients and sporadic nasopharyngeal carcinoma patients and normal control group S100A9 mRNA expression were respectively-3.886±1.663,-3.421±1.110,-4.031±1.586,the comparison of three group P>0.05,the difference hadn't statistically significant.The protein expression of S100A9 was 0.518±0.353,0.481±0.344,and 0.245±0.202,respectively,and the difference was statistically significant(P<0.05).Conclusion S100A9 is differentially expressed of peripheral blood lymphocytes in high familial nasopharyngeal carcinoma,sporadic nasopharyngeal carcinoma patients and normal people.S100A9 is expected to be an early screening indicator for nasopharyngeal carcinoma.
