摘要
目的探究白藜芦醇是否通过SATB2调控HCT 116细胞凋亡及其机制。方法用不同浓度白藜芦醇(resveratrol,Res)处理HCT 116细胞24 h,确定最佳Res给药量为8.33μg/mL。HCT 116细胞分为control组、si-SATB2组、si-SATB2+Res组、空质粒组、SATB2过表达组、Res组、SATB2过表达+Res组,处理结束后,收集细胞提取总蛋白,Western blot检测SATB2、Bax、Bcl-2,Caspase-3、Caspase-9,Cyt-C蛋白的表达。结果 (1)Res上调SATB2蛋白的表达。(2)抑制SATB2表达、开高Bcl-2/Bax比值,下调Cyt-C、Caspase-3、Caspase-9蛋白的表达;Res可逆转此作用。(3)过表达SATB2降低Bcl-2/Bax比值,上调Cyt-C、Caspase-3、Caspase-9蛋白的表达,Res加强此作用。结论抑制SATB2降低HCT 116细胞凋亡水平,Res能够通过上调SATB2促进HCT 116细胞Bax-Caspase3、9凋亡通路蛋白表达。
Objective To investigate whether resveratrol regulates apoptosis of HCT 116 cells through SATB2 and its mechanism.Methods HCT 116 cells were treated with resveratrol(Res)at different concentrations for 24 h,and the optimal dose of Res was determined to be 8.33μg/mL.HCT 116 cells were divided into control group,si-SATB2 group,si-SATB2+Res group,empty plasmid group,SATB2 overexpression group,Res group,and SATB2 overexpression+Res group.After treatment,total protein was extracted from the cells,and the protein expressions of SATB2,Bax,Bcl2,Caspase-3,9 and Cyt-C were detected by Western Blot.Results (1)Resveratrol up-regulated the expression of SATB2 protein in HCT116 cells.(2)Knocking down of SATB2 gene incveased the Bcl-2/Bax ratio and down-reglated the expression of CYT-C,Caspase-3 and Caspase-9 proteins,and Res reversed these effects.(3)Overexpression of SATB2 reduced the Bcl-2/Bax ratio in HCT 116 cells and down-regulate the expression of Cyt-C,Caspase-3 and Caspase-9 proteins,while Res enforced these process.Conclusion Inhibition of SATB2 reduced the level of apoptosis in HCT 116 cells,and Res could promote the expression of apoptotic protein in HCT 116 cells by up-regulating SATB2.
