摘要 为研究黄花蒿青蒿素提取工艺及其抑菌活性,采用正交试验优化回流提取法提取青蒿素的工艺条件,并用滤纸片扩散法和倍比稀释法测定青蒿素对金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、酿酒酵母和黑曲霉的抑菌活性。结果表明,回流提取法提取青蒿素各因素主次顺序为乙醇浓度>萃取温度>固液比>萃取时间,最佳生产工艺参数为浓度65%、固液比1:8、提取温度80℃、提取时间4h。此条件下的青蒿素对金黄色葡萄球菌、枯草芽孢杆菌、大肠埃希菌、酿酒酵母和黑曲霉均有一定抑菌活性,且对金黄色葡萄球菌和酿酒酵母的抑菌效果最好,最小抑菌浓度(MIC)为48.8mL/L。 In order to study the extraction technology and antibacterial activity of artemisinin from Artemisia annua,the orthogonal experiment was used to optimize the reflux extraction process of artemisinin, and the antibacterial activity of artemisinin against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Saccharomyces cerevisiae and Aspergillus niger was determined by filter disk diffusion method and multiple dilution method. The results showed that the order of reflux extraction was ethanol concentration > extraction temperature > solid-liquid ratio >extraction time. The optimal production parameters are the concentration of 65%, the solid-liquid ratio of 1∶ 8, the extraction temperature of 80 ℃, the extraction time of 4 h. Under the conditions, artemisinin had certain antibacterial activity against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Saccharomyces cerevisiae and Aspergillus niger, and had the best antibacterial effect on Staphylococcus aureus and Saccharomyces cerevisiae, with MIC value of 48.8 mL/L.
