Box-Behnken响应面法优化西藏三颗针中盐酸小檗碱的提取工艺

Optimization of extraction process of berberine hydrochloride from Berberis diaphana Maxim in Tibet by Box-Behnken response surface methodology

优化西藏三颗针中盐酸小檗碱的提取工艺。采用比色法测定盐酸小檗碱的含量,以盐酸小檗碱含量为指标,分别考察pH值、稀酸浓度、溶剂温度、料液比四因素对提取分离盐酸小檗碱的影响,并结合Box-Behnken响应面分析法,优化西藏三颗针中盐酸小檗碱的提取工艺。结果表明,最佳提取工艺稀酸浓度为0.2%,pH值为10,提取温度50℃,料液比为1∶8时,该工艺下盐酸小檗碱的得率最高。说明优化后的提取工艺简便可行,收率高,具有工业化生产推广价值。

To optimize the extraction process of berberine hydrochloride DC from three needles of Xizang Province. Using colorimetric method to determine the content of berberine hydrochloride,the content of berberine hydrochloride as indicators,respectively to investigate concentration of lime milk adjust pH value,dilute acid concentration,solvent,temperature,materialliquid than four factors on the extraction and separation of berberine hydrochloride DC,and combined with the Box-Behnken response surface analysis,optimization of Tibet three needle in the extraction process of berberine hydrochloride. The results showed that the optimal extraction conditions were as follows:dilute acid concentration of 0.2%,adjusted p H value of lime milk concentration of 10,extraction temperature of 50 ℃,solid-liquid ratio of 1 ∶8,the extraction yield of berberine hydrochloride was the highest. The results showed that the optimized extraction process was simple,feasible,high yield,and had the value of industrial production and popularization.
