四氢姜黄素对光老化小鼠皮肤的修复作用 被引量:1

Repairing Effect of Tetrahydrocurcumin on the Skin of Photoaging Mice

摘要 目的:探讨四氢姜黄素对紫外线(UVA+UVB)辐射致小鼠皮肤光老化的保护作用。方法:KM雌性小鼠随机分为正常对照组、模型对照组、四氢姜黄素100 mg/kg组,每组10只。模型对照组与四氢姜黄素100 mg/kg组采用最小红斑剂量(100 mJ/cm^(2))递增法进行紫外线照射。实验过程中,四氢姜黄素100 mg/kg组每日给予四氢姜黄素灌胃,正常对照组、模型对照组每日口服0.5%羟甲基纤维素钠,实验持续十周。实验结束24 h后,评价小鼠皮肤外观,评价结束后将小鼠断颈处死,取背部皮肤,观察其病理学变化,并对超氧化物歧化酶(SOD)活力、羟脯氨酸(Hyp)含量进行测定。结果:与正常对照组比较,模型对照组皮肤皱纹明显加重,出现皮革样改变、红肿破溃,超氧化物歧化酶(SOD)活力显著降低(P<0.01),羟脯氨酸(Hyp)含量显著降低(P<0.01);与模型对照组比较,四氢姜黄素100 mg/kg组皮肤皱纹减轻,超氧化物歧化酶(SOD)活力显著增高(P<0.01),羟脯氨酸(Hyp)含量显著增高(P<0.01)。结论:四氢姜黄素对光老化小鼠皮肤具有一定的修复作用,可能与抑制氧化应激有关。 Objective:To explore the protective effect of tetrahydrocurcumin on the photoaging of mice skin induced by ultraviolet(UVA+UVB)radiation.Methods:KM female mice were randomly divided into the control group,the model control group,and 100 mg/kg tetrahydrocurcumin group,with 10 mice in each group.The mice in the model control group and 100 mg/kg tetrahydrocurcumin group were irradiated with ultraviolet rays according to the method of increasing the minimum erythema dose(MED=100 mJ/cm^(2))obtained in the experiment.During the experiment,mice in 100 mg/kg tetrahydrocurcumin group was orally administered with tetrahydrocurcumin daily,and 0.5%sodium hydroxymethylcellulose was given daily to mice in the control group and model control group,for ten weeks.24 hours after the end of the experiment,the appearance of the mouse skin was evaluated.After the evaluation,the mice were sacrificed by cervical dislocation.The skin on the back was taken to observe the pathological changes,the activity of superoxide dismutase(SOD)and the content of hydroxyproline(Hyp)were determined.Results:Compared with the control group,the mice in the model group had significantly worse skin wrinkles,leather-like changes,redness and swelling,the SOD activity was significantly reduced(P<0.01),and Hyp content was significant decreased(P<0.01).Compared with the model group,100 mg/kg tetrahydrocurcumin group reduced skin wrinkles,the SOD activity was significantly increased(P<0.01),and hydroxyproline(Hyp)content was significantly increased(P<0.01).Conclusion:Tetrahydrocurcumin has a certain repair effect on the skin of photoaging mice,which may be related to the inhibition of oxidative stress.
