摘要
目的:探究葛根、粉葛对异丙肾上腺素诱导的大鼠心肌缺血的影响。方法:本实验通过注射异丙肾上腺素诱导大鼠心肌缺血的发生,分别灌胃维拉帕米4 mg/kg、葛根3.15、1.58、0.79 g/kg、粉葛3.15、1.58、0.79 g/kg,连续20 d,考察葛根与粉葛对心肌缺血大鼠Ⅱ导联心电图参数,大鼠血清中谷草转氨酶(AST)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、肌钙蛋白T(cTnT)、超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子(TNF-α)的含量以及大鼠心肌组织病理学变化。结果:与正常对照组相比,模型对照组的|ΔJ|值、心率、大鼠血清中的AST、LDH、CK、cTnT含量明显升高(P<0.05或P<0.01);MDA含量显著升高(P<0.01);心肌组织结构排列紊乱,心肌细胞间隙增大,心肌组织充血,心肌纤维断裂严重,有炎症发生;与模型对照组相比,维拉帕米4 mg/kg、葛根3.15、1.58、0.79 g/kg组、粉葛3.15、1.58、0.79 g/kg组的|ΔJ|值、AST、LDH、CK、cTnT含量明显降低(P<0.05或P<0.01),血清中SOD活性明显升高(P<0.05或P<0.01),维拉帕米4 mg/kg、葛根0.79 g/kg、粉葛0.79 g/kg的MDA含量明显下降(P<0.05或P<0.01),维拉帕米4 mg/kg、粉葛0.79 g/kg组的TNF-α含量显著下降(P<0.05);心肌组织结构排列较整齐,改善心肌纤维断裂情况,炎症反应减轻,组织充血情况改善。结论:葛根、粉葛均对异丙肾上腺素注射液所致的心肌缺血有保护作用且粉葛药用价值不低于葛根,其作用机制可能与抗氧化有关。
Objective:To investigate the effects of Puerariae Lobatae Radix and Puerariae thomsonii radix on isoproterenol-induced myocardial ischemia in rats. Methods:In this experiment, the myocardial ischemia in rats was induced by injection of isoproterenol, then 4 mg/kg Verapamil, 3.15 g/kg, 1.58 g/kg and 0.79 g/kg Puerariae lobatae radix, 3.15 g/kg, 1.58 g/kg and 0.79 g/kg Puerariae thomsonii radix were orally administered for 20 days. Then parameters of electrocardiogram(ECG) in rats with myocardial ischemia were detected, the contents of glutamic oxaloacetic transaminase(AST), lactate dehydrogenase(LDH), creatine kinase(CK), troponin T(cTnT), superoxide dismutase(SOD), malondialdehyde(MDA) and tumor necrosis factor(TNF-α) were determined, the myocardial histopathology was observed. Results: Compared with the control group, The |ΔJ| was significantly increased in the model group, the heart rate and the contents of AST、LDH、CK and cTnT were significantly increased(P<0.01 or P<0.05), then MDA content was significantly increased(P<0.01), while the myocardial histological arrangement was in disorder, myocardial interstitial space was enlarged, myocardial tissue was congested, myocardial fiber was severely ruptured, and the inflammation occurred. Compared with the model group, the |ΔJ| value and the contents of AST、LDH、CK and cTnT were significantly decreased in 4 mg/kg Verapamil, 3.15 g/kg, 1.58 g/kg and 0.79 g/kg Puerariae lobatae radix groups, 3.15 g/kg, 1.58 g/kg and 0.79 g/kg Puerariae thomsonii radix groups were significantly decreased(P<0.01 or P<0.05), while the serum SOD activity was significantly increased(P<0.01 or P<0.05), The MDA content was significantly decreased in 4 mg/kg Verapamil group, 0.79 g/kg Puerariae lobatae radix group and 0.79 g/kg Puerariae thomsonii radix group(P<0.01 or P<0.05), The content of TNF-α was significantly decreased in 4 mg/kg Verapamil group and 0.79 g/kg Puerariae thomsonii radix group(P<0.01 or P<0.05). The structure of myocardial tissue was arranged in order, the rupture of myocardial fiber was improved, the inflammatory reaction and tissue congestion were alleviated.Conclusion:Puerariae lobatae radix and Puerariae thomsonii radix have the protective effect on myocardial ischemia induced by isoproterenol injection, the medicinal value of Puerariae thomsonii radix is not lower than that of Puerariae lobatae radix, and its mechanism may be related to antioxidation.
