摘要
对显微切片技术在枇杷研究中的应用进行综述。总结了徒手切片在观察枇杷构造上的简单应用,石蜡切片在观察枇杷花芽分化、胚胎发生、果实及叶片组织上的应用,超薄切片在观察胚胎发生的细胞结构和果肉细胞质体超微结构上的应用,以及冰冻切片在研究果肉细胞壁组分空间分布的动态变化和种间杂种及其亲本果实的比较解剖学方面的应用。通过比较4种切片技术的优缺点,提出:4种技术在枇杷研究中有各自的适用范围,研究者需根据研究目的和材料特点,选择合适的切片技术或同时应用2种甚至3种技术,才能将它们更好地应用于观察乃至解释枇杷的显微结构。
In this paper,we review the application of four microsectioning techniques in the research on loquat.First of all,we introduce the application of freehand sectioning in observing the structure of loquat.Secondly,we discuss the contribution of paraffin sectioning to the observation of flower bud differentiation,embryogenesis and the tissues of loquat fruits and leaves.Thirdly,we examine the great help of ultrathin sectioning to the observation of the subcellular structure during the period of embryogenesis and the ultrastructure of cytoplasmic loquat pulps.Lastly,we discuss the application of frozen sectioning which contributes to the study on dynamic changes in the spatial distribution of cell wall components in loquat pulps and the comparative anatomy of interspecific hybrids and their parents.By comparing the advantages and disadvantages of those four sectioning techniques,we propose that each technique has its own scope of application in the observation of the internal structure of loquat.Based on different research purposes and material characteristics,researchers can select appropriate microsectioning techniques or apply more than two microsectioning techniques so as to better observe and explain the microstructure of loquat.
