摘要
目的研究香烟烟雾颗粒对大鼠肠系膜动脉肾上腺素α_(1)受体的作用及机制。方法采用离体器官培养技术,离体肌张力描记观察苯肾上腺素引起的平滑肌收缩量效曲线变化,考察香烟烟雾诱导肾上腺素α_(1)受体在血管高反应性中的变化及调节肾上腺素α_(1)受体反应的相关信号通路。结果香烟烟雾可以增强肾上腺素α_(1)受体激动剂引起的血管收缩,ERK1/2通路抑制剂可减轻血管收缩,降低肾上腺素α_(1)受体的表达。NF-κB信号通路抑制剂和AMPK信号通路激活剂对香烟烟雾颗粒引起的肾上腺素α_(1)受体介导的血管收缩没有影响。结论香烟烟雾通过ERK1/2信号通路上调肾上腺素α_(1)受体介导的血管收缩。
Objective To determine the effect of cigarette smoking particles on the vasoconstrictions induced byα_(1)-adrenoceptor in the mesenteric artery in rats.Methods In vitro organ culture was used to determine how cigarette smoke particles affected the vasoconstrictions induced byα_(1)-adrenoceptor and its mechanisms in vascular smooth muscle cells by myograph.Results Cigarette smoke particles enhanced the mesenteric artery constriction induced byα_(1)-adrenoceptor agonist.The inhibitors of ERK1/2 signaling pathway alleviated the vasocon striction and decreased the protein expression ofα_(1)-adrenoceptors.However,NF-κB signaling pathway inhibitors and AMPK signaling pathway activators had no effect on theα_(1)-adrenoceptor mediated vasoconstriction induced by cigarette smoke particles.Conclusion Cigarette smoking particles can enhance the vasoconstrictions mediated byα_(1)-adrenoceptor through ERK1/2 signaling pathway.
